| Fructus Aurantii is a kind of dried and immature fruit belonging to Citrus aurantium L. and its cultivated variety. Also it is one of geo-authentic crude drugs for Jiangxi. This topic made a systematic research about chemical compositions, processing technology,and the main pharmacodynamics experiments after processing on Fructus Aurantii according to Zhang-Band processing technology in Jiangxi traditional school of Fructus Aurantii. And it compared with the processed products of Fructus Aurantii Pharmacopoeia Method. The main researches are the followings:1.Processed Technology Research:Processing methods of Fructus Aurantii have been systematically studied such as cutting.The dried technology has been optimized by the indexes of neohesperidin, naringin, moisture and the volatile oil. There has been optimized by the indexes of neohesperidin and naringin,mellow dissolved substance,the volatile oil and properties in the fried by wheat bran technology. TLC has been established through the standard reference of naringin and neohesperidin in the Fructus Aurantii.We has also instituted TLC of the wheat bran by the standard reference of leucine,aminovaleic acid and aminopropionic acid.General experimental results come to the conclusion that the best cutting specific-ations is the Chinese herbal piece of 1.5 mm; optimum drying temperature is 60℃, time is 60min; optimum fried by wheat bran temperature is 190℃, time is 150s, added the volume is 15% wheat bran. Established the best process Technology of Chinese h-erbal piece of Fructus Aurantii.2. Pharmacodynamic Study of the Chinese herbal piece of Fructus Aurantii: Fructus Aurantii on this topic on the gastrointestinal aspects of the kinetics study are as follows:(1) Fructus Aurantii and its processed products producing the impact of intestinal propulsion in mice:We have proceeded intestinal promote in normal mice with the crude Fructus Aurantii, product fried by wheat bran in Pharmacopoeia, crude product in Zhang-Band, product fried by wheat bran in Zhang-Band.The results show that Fructus Aurantii and its processed products are able to strengthen intestinal propulsive function in mice,there are significant difference compared with the distilled water group. The product fried by wheat bran in Pharmacopoeia and the product in Zhang-Band are more relaxatisive than the crude product in promoting mouse intestinal propulsive function,and reported in the literature "Fructus Aurantii fried by wheat bran to ease its fierce and free of injuring vigor."and " Processed by wheat bran,restraining its fierce and not on diaphragm injury"consistent with the theory.(2) Study of about the Mechanism about the intestinal propulsion in mice with various ingredients of Fructus Aurantii:We have studied the influence of intestinal propulsion in mice with the volatile oil, water-soluble ingredients, fat-soluble components in Fructus Aurantii,and to explore the intestinal propulsive mechanism of above-mentioned components in mice. The results showed that the 10% volatile oil group, the 20% volatile oil group, four times the volume of water-soluble components group, eigth times the volume of fat-soluble components group can strengthen intestinal propulsive function, and there are significant different compared with the distilled water group,20% volatile oil have antagonistic effects about the inhibition of mouse small intestine propulsion caused by adrenaline and diphenhydramine; water-soluble components have antagonistic effects about the inhibition of mouse small intestine propulsion caused by adrenaline; the fat-soluble components have antagonistic effects about the inhibition of mouse small intestine propulsion caused by atropine,diphenhydramine;and can enhance the small intestine propulsive hyperfunction effects caused by neostigmine.The results show that the volatile oil promoting the intestine propulsion may be related to adrenergic system and histamine Hi-R; the water-soluble components promoting the intestine propulsion may be related to adrenergic system; and the fat-soluble components promoting the intestine propulsion may be related to cholinergic M-R, H1-R and cholinesterase.(3) The effects of isolated rabbit small intestine with the Chinese herbal piece of Fructus Aurantii and its volatile oil:We have studied the influence of isolated rabbit intestine with the crude prude,Fructus Aurantii fried by wheat bran and volatile oil.The results showed that, decoction and volatile oil can restrain the spontaneous activity on isolated rabbit intestinal smooth muscle,with the dosage increased and strengthened,is a significantly dose-effect relationship;the product fried with wheat bran is more mitigated than the crude product.(4) The effects of Gas and MTL in rat blood with the Chinese herbal piece of Fructus Aurantii:We have studied the contents of Gas and MTL in the rat blood with the crude product, processed products by pharmacopoeia, processed products by Zhang-Band.The results show that Fructus Aurantii and its processed products can raise the contents of Gas and MTL in blood, compared with the distilled water group,there is significant difference;It notes that Fructus Aurantii function not only promote the intestinal propulsion in the gastrointestinal tract by receptor,but also can stimulate parts of the gastrointestinal cells Gas, MTL, so that endogenous Gas, MTL levels promote the propulsion of gastrointestinal function and secretion.To sum up, under the guidance of traditional Chinese medicine theory, this paper applied the combination of traditional and modern methods, studied systematically the chemical composition of Fructus Aurantii processed by Zhang-Band producing the traditional area of Jiangxi,optimized the processing technology,analyzed the component of pre-processed and post-processed,researched the fingerprint of different habitats.Finally,we have also proceeded the pharmacodynamics experiments-preliminary study of gastrointestinal kinetics.The scientific basis is provided in order to achieve the processing technology normalization and standardization of the Chinese herbal piece of Fructus Aurantii. |
