Objective: To measure the level of serum ET-1and NO, the expression of PAI-1 and t-PA in diabetic rats thoracic aorta tissue after Furongtongmai capsule treatment,and explore the possible anti-atherosclerosis mechanism of Furongtongmai capsules.Methods: 10 of the 60 adult male Wistar rats were randomly chosen as blank control group(NC, n = 10), and the remaining formed diabetes model groups. The other 50 rats were injected STZ 60 mg/kg one-time on the lower enterocoelia to copy diabetes model,and the control group rats were injected with the same amount of sodium citrate buffer. After 72 hours,the rats whose blood glucose exceeded 16.7mmol/L were identified as diabetic model rats.40 of 50 diabetes rats were randomly divided into the follow groups:DM(n=10), FRL(n=10),FRM(n=10),FRH(n=10). FRL group was given 0.7g/(kg body weight) to fill the stomach, FRM group was given the 1.4 g/(kg body weight), FRH grou-p was given 2.8 g/(kg body weight), the rest of the two groups of daily to give the same dose of normal saline. During the whole experiment, each rat freely ate and drank water. After 8 weeks,we gave all the rats a general anaesthetic with 3% pentobarbital(40 mg/kg),quickly took thoracic aortic blood to measure BS TG,TC,NO,ET-1.Then we observed the expression level of PAI-1,t-PA with immunohisto- chemical technique.The Measurement data were showed with mean±standard deviation.We used single factor analysis of variance for two groups comparisons,.K-W-H tests if necessary, paired t tests for the comparisons before and after treatment.All the datas were processed with SPSS 16.0statistical software.The inspection level: α=0.05.1 The influence on the general condition of rats. The rats of DM 、FRL、FRM、FRH groups ate more food,drank more water and were less active than the NCgroup.Their hairs color were darker than the NC group rats..2 The effect on the blood sugar(BS) Before the treatment, the BS of DM 、FRL、FRM、FRH groups was higer than NC group(P < 0.05); the BS of DM 、FRL、FRM、FRH groups had no statistical difference P > 0.05).After treatment, the BS of DM、 FRL、FRM、FRH groups were higer than NC group(P<0.05);the BS of DM 、FRL、FRM、FRH groups had no statistical difference(P > 0.05).There were no statistical difference before and after the treatment among every group rats(P > 0.05).3 The change of serum ET-1 Compared with NC group,the ET-1 of the DM、FRL、FRM、FRH groups increased obviously(P< 0.05). Compared with DM group,the change of the FRL group had no statistical differrence(P > 0.05), the ET-1 of FRM、FRH groups decreased(P < 0.05).Compared with FRL group,t he ET-1 of FRM、FRH groups decreased(P<0.05). Compared with FRM group,t he change of FRH group had no statistical differrence(P> 0.05).4 The impact on serum NO Compared with NC group,the NO of the DM 、 FRL 、 FRM 、 FRH groups decreased obviously(P < 0.05).Compared with DM group,the change of the FRL group had no statistical differrence(P > 0.05), the NO of FRM 、 FRH groups increased(P< 0.05).Compared with FRL group,t he ET-1 of FRM、FRH groups increased(P <0.05). Compared with FRM group,t he change of FRH group had no statistical differrence(P > 0.05).5 The expression of PAI-1 in thoracic aortic.The momal tissue had little PAI-1 expression. Compared with NC group,the expression of the DM、FRL、FRM、FRH groups increased obviously(P < 0.05). Compared with DM group,the change of the FRL group had no statistical differrence(P > 0.05), the PAI-1expression of FRM、FRH groups decreased(P< 0.05). Compared with FRL group,t he expression of FRM 、FRH groups Results:decreased(P < 0.05). Compared with FRM group,t he change of FRH group had no statistical differrence(P > 0.05).6 The expression of t-PA in thoracic aortic. The momal tissue had much PAI-1 expression. Compared with FRL group,the expression of FRM、FRH groups increased(P< 0.05).Compared with FRM group, the change of FRH group had no statistical differrence(P > 0.05).Conclusion:1 After eight- week-experiment,we succesfully copied the diabetic rats models.2 Furongtongmai capsule had no obvious effect on diabetic rats glycometabolism.3 Furongtongmai capsule could decrase the serum ET-1 and raise the level of NO in diabetes rats.4 Furongtongmai capsule could improve the expression of t-PA,inhibit the over-expression of PAI-1 in thoracic aorta tissues..5 The experiments show that Furongtongmai capsule may prevent AS from progressing through protecting vascular endothelial cells and regulating the fibrinolytic system. Middle dose is enough when we treat diabetic macroangiopathy.
