Objective: Copy diabetic rats models, observed the complex Furongtongmai capsule on diabetic rats kidney PDGF- BB and the expression of MMP- 9, discusses the Furongtongmai capsule in the treatment of diabetic nephropathy mechanism, provide reliable test for the Chinese medicine treatment of diabetic nephropathy.Methods: Wistar rats(male adult health, n = 60), the rats were randomly divided into normal control group(NC group, n = 10) and module 50 rats.Build module fasting 12 hours later, the rats abdominal cavity of disposable injection chain urea with cephalosporins(STZ) 60 mg/kg(STZ before use with 0.1 M with the concentration of 1% sodium citrate buffer solution, pH =4.2) diabetic rats model was established, by intraperitoneal injection of NC the same amount of sodium citrate buffer. After 72 hours, end of peripheral venous blood test blood sugar, blood sugar levels or greater tendency for16.7mmol/L is determined for diabetic rats. Building after a successful rats randomly selected 40, divided into diabetic model group(DM, n = 10), the Furongtongmai capsule low-dose treatment group(FRL, n = 10), the Furongtongmai capsule dose in the treatment group(FRM, n = 10), the Furongtongmai capsule high dose treatment group(FRH, n = 10). After a week, according to the human body- the table surface area ratio of rats conversion equivalent dose in rats, for Furongtongmai dose group was given in 1.4 g/kg(? d) to fill the stomach, the Furongtongmai low dose group was given 0.7 g/(kg, d) to fill the stomach, the Furongtongmai high dose group was given 2.8 g/kg(? d) to fill the stomach, gastric 1 times a day, eight weeks in a row. Normal control group and diabetic model group daily to give equal dose of normal saline lavage. All free diet water of rats during experimentalobservation, forage standards, the environment temperature is 22 ℃- 26 ℃,the experiment is not used during the insulin and blood sugar(or) other oral glucose-lowering drugs intervention. Lavage treatment for 8 weeks, caudal vein blood glucose measurement, with 3% pentobarbital sodium 80 mg/kg intraperitoneal injection of anesthetic rats, midline incision abdomen,abdominal aortic blood, blood samples. Take blood after harvesting the rat kidney, removing renal capsule and the renal hilum connective tissue and physiological saline flushing after weighing, will be recorded in the kidney is less than 0.5 cm x 0.5 cm x 0.1 cm tissue block, at 4% of sex in formaldehyde solution fixed, HE staining under light microscopy observation of glomerular and renal tubular interstitial pathological changes, immunohistochemical SP method to observe the pathological tissue positive staining in the area and intensity.All the data with SPSS 16.0 statistical software processing, inspection level:α=0.05. Measurement data with——X ±SD said. Variance together,compares the single factor analysis of variance between groups, group comparison before and after the treatment using matching t test.Results:1 Building groups before the rats’ blood sugar has no statistical significance(P > 0.05), compared with before treatment, compared with NC group, DM group and FRL, FRM, FRH group rats blood glucose were significantly higher(P< 0.05), while the DM group and FRL, FRM, FRH blood sugar is no statistical significance between groups(P> 0.05). After lavage treatment for 8 weeks, compared with NC group, DM group and FRL,FRM, FRH group blood sugar of rats, were significantly increased(P< 0.05),while the DM group and FRL, FRM, FRH group rats blood glucose is no statistical significance(P > 0.05); Groups of rats blood glucose compared with before treatment has no obvious difference(P > 0.05).2 Lavage treatment after 8 weeks, compared with NC group, DM group and FRL, FRM, FRH TG, TC level increased significantly(P < 0.05); DM group, FRL group, FRM and FRH way comparison between TG, TC level noobvious difference(P > 0.05).3 Lavage treatment after 8 weeks, compared with NC group, DM group and FRL, FRM, FRH Scr and BUN level increased significantly(P < 0.05);Group compared with DM group, FRL Scr and BUN level without obvious difference(P > 0.05), FRM, FRH group of Scr and BUN levels drop significantly, with statistical significance(P<0.05); Compared with FRL group, Scr, BUN level FRM, FRH group was obviously lower(P < 0.05);FRM, FRH comparison between groups, Scr, BUN level no obvious difference(P > 0.05).4 PDGF- BB expressed in normal rat kidney has a small, compared with NC group, DM group and FRL, FRM, FRH PDGF- BB expression increased significantly(P < 0.05); Group compared with DM group FRL, FRM, FRH PDGF- BB expression significantly decreased(P < 0.05); Compared with group FRL FRM PDGF- BB expression, FRH group was obviously lower(P< 0.05); FRM, FRH group comparison PDGF- BB expression has no obvious difference(P> 0.05). MMP-9 in normal rat renal normal expression, compared with NC group, DM group and FRL expression of MMP- 9, FRM, FRH group significantly decreased(P<0.05); Group compared with DM group FRL,FRM, FRH MMP-9 expression increased significantly(P < 0.05); Compared with FRL group, MMP-9 express FRM, FRH group was obviously increased(P < 0.05); Comparison between FRM, FRH group, no difference between the expression of MMP-9(P> 0.05).Conclusion:1 Furongtongmai capsule does not reduce the glucose and lipid metabolism disorder.2 Furongtongmai capsule can reduce the renal hypertrophy in diabetic rats and lower blood urea-nitrogen, serum creatinine. After Furongtongmai capsule can inhibit the increase of serum CRP, inhibit the rise of PDGF- BB expression, can inhibit renal expression of MMP-9 down.3 This study show Furongtongmai capsule probably through above mechanisms to delay of renal injury in diabetic rats.
