Diabetes mellitus is a kind of whole body chronic metabolic disease with high glucose as the main biochemical characteristics.Chronic complications are the leading cause of death and disability in diabetes,especially macroangiopathy.So study the pathogenesis of diabetic macroangiopathy and looking for the prevention and treatment of diabetic macroangiopathy of drug targets,to improve the quality of life of patients with diabetes and prolong life is particularly important.Several studies have clearly shown the small guanosine-5-triphosphate-binding protein RhoA and its downstream target Rho-kinase induced the actin cytoskeletal assembly,regulation of gene trascription and cell cycle after it act on the cytoskeletal and its target proterin.The Rho/ROCK pathway participates in a variety of important pathophysiological functions in vasculature including SMC contraction,cell migration,cell adhesion,and inflammatory cell responses,as well as gene expression.This article reviews the biologial characteristics of the Rho/ROCK pathway and the relationship between it and the development or progression of diabetic macroangiopathy to provide a novel and fruitful strategy for the prevention and treatment of diabetic macroangiopathy.
