Objective:Immunohistochemistry double staining technique was used to observe the changes of myoepithelial cells in the rat atrophy parotid gland.Methods:Ligating the tube by the side of the rat parotid gland in order to promote the glands atrophy. The rats were put to death after the ligation of 1,3,5,7,14,21,30,60, 90,120,90,120 days, then gathered the atrophic parotid gland specimens of different times. Immunohistochemical double staining and IHC double notation was performed to quantitative analyse the changes in number, proliferation rate and distribution of myoepithelial cells during atrophy of the ratparotid gland.Results:Apoptosis of acinar cells and formation of a large number of duct-like structures, fibrous tissue hyperplasia and inflammatory cells infiltration after ligation. Quantitative analysis of immunohistochemical staining results showed that:on the third day after duct ligation, the myoepithelial cells proliferated rapidly; the proliferation rate reached a peak value on the fifth day and then decreased; on the 30d-150d, the proliferation rate were maintained at a low level; It is difficult to observe the proliferation of myoepithelial cell and the myoepithelial cell number was significantly decreased.Conclusion:Myoepithelial cells proliferated rapidly up to day 5 after ligation, then followed by gradual increase at a low level, at last it was followed by a rapid decrease after the total number reached the peak in 180 days.
