Research Of The Relationship Betreen MiRNA181a And Treg Cell In Peripheral Blood Of Primary Immune Thrombocytopenia

Objective:Immune thrombocytopenia(ITP) is an autoimmune disease with T lymphocyte subsets imbalance,characterized by increased bleeding tendency 。 The reason of T lymphocytel subsets imbalance in ITP is not known,but treg cells can clearly be involved in the pathogenesis of ITP。It is play an important role in ITP。Because mi RNA181 a has an important role in T cell differentiation and mi RNA181 a expression in peripheral blood T lymphocytes was significantly reduced, mi RNA181 a may be involved in the ITP T lymphocyte subsets imbalance and treg physiological processes of cells.Through the gene expression of mi RNA181 a 、treg cell and TGF-beta,we will observe the correlatiao of the mi RNA181 a expression whit treg cell and discuss and their relatiaonship in the pathogenesis of ITP patients.Method:1,the separation peripheral blood T lymphocytes.2,the level of the gene expression of mi RNA181 a by pt PCR.3,the level of treg cell by flow cytometry.4,the level of TGF-β levels in plasma by Elisa.5,SPSS statistical analysis software was used for statistical analysis,and to analysis the correlatiao of the mi RNA181 a expression whit treg cell.Results:1.The expression of peripheral blood T lymphocytes m RNA181 a of:ITP group: 0.495 ± 0.109 normal group: 1.296 ± 0.450.ITP group was significantly lower than the normal group, P <0.001 was considered statistically significant.2. The peripheral blood CD4 + T lymphocytes in Treg cells(CD4+ Foxp3) ratio: ITP group: 1.82 ± 0.46. Normal group: 7.21 ±1.08 ITP group was significantly lower than the normal group,P <0.05, the difference was statistically significant.T lymphocytes in the peripheral blood plasma TGF-β levels in the peripheral blood of 3: ITP Group: 3211.21 ± 1100.11 normal group: 3211.21 ± 1100.11. ITP group was significantly lower than the normal group, P <0.05, the difference was statistically significant.micro RNA181 a and treg cell correlation analysis: micro RNA181 a expression and treg cell ratio was positively correlated, P<0.05, r = 0.99.Conclusion1, the mi RNA181 a expression si lower in ITP patient.2,T lymphocyte immune function on treg cell in ITP patient was in the inhibitory state3, m RNA181 a expression in ITP patients was positively correlated with the proportion of Treg cells.4, m RNA181 a expression may be involved whit treg cell disorders in the ITP.
