Objective:According to different stages and types,we divided those vitiligo patients as different groups,and compared with healthy controls to observe the changing of Th22,Th17,Treg cells related cytokines levels in different groups,preliminarily discussed Th22,Th17,Treg cells’ role in the development of vitiligo mechanism by comparing with IL-22 m RNA,IL-17 m RNA,Foxp3 m RNA in PBMC,and serum IL-22,IL-17,Foxp3 protein expression level;According to different types and stages,preliminary discussed the effects and mechanism of Th22,Th17 and Treg cells in on the development of vitiligo:Summarize the relationship of three kinds of cells related cytokines in vitiligo patients’ peripheral blood,and make sure that the pathogenesis of vitiligofurther so clear that to assess the development and prognosis of vitiligo,provide a reference for the treatment of vitiligo.Methods:We collected a total of 60 cases of patients with vitiligo diseases from October 2015 to October 2016 in department of dermatology,Affiliated hospital of tianjin academy of traditional Chinese medicine,according to different stages,we divided two groups which include 30 case of active patients,30 cases of stationary patients;According to different types,we divided 29 cases of ordinary patients and31 cases of segmental patients.We selected 30 cases of healthy people as normal control,whose age and sex had matched.Detailedly recorded patients’ name,gender,age,and invasion site,body surface area,the fixed number of year and family history,etc.Routinely separated peripheral blood mononuclear cells(PBMC),and used Real-Time RT-PCR detect the levels of IL-22 m RNA,IL-17 m RNA,Foxp3 m RNA expression in PBMC;Taking peripheral venous blood,double antibody sandwich ELISA detected the levels of peripheral blood IL-22,IL-17,Foxp3.In order to make clear Th22 expression in vitiligo,we made those test results associated with vitiligo activity analysis,and analysed the correlation between the three by statistical regressive method to clarify Th22,Th17,Treg cells in vitiligo.Using statistical methods for the analyse experimental results,P<0.05.Results:1.In the experimental group,Vitiligo patients’ IL-22 m RNA,IL-17 m RNA and Foxp3m RNA level in PBMC and IL-22,IL-17,Foxp3 protein are significantly higher than healthy controls,and the activity of vitiligo patients is higher than vitiligo patients who were in stable stage.On the other hand,in the experimental group,Foxp3 m RNA level in PBMC and Foxp3 protein are lower than healthy controls,and the activity of vitiligo patients is higher than vitiligo patients who were in stable stage The difference had statistical significance.2.Ordinary vitiligo patients’ IL-22 m RNA,IL-17 m RNA levels in PBMC and IL-22,IL-17 protein levels in serum were significantly higher than segmental vitiligo patients;On the contrary,ordinary vitiligo patients’ Foxp3 m RNA level in PBMC and Foxp3 protein level is lower than segmental vitiligo patients.3.In vitiligo patients’ peripheral blood:IL-22 m RNA,IL-17 m RNA level in PBMC and IL-22,IL-17 protein in serum have dirert proportional correlation,both of them are inversely proportional to Foxp3 m RNA level in PBMC and Foxp3 protein level.Conclusions:1.In vitiligo patients’ peripheral blood,Th22,Th17 cells related factor expression and translation level were higher than healthy controls,and active patients is higher than stationary patients.This illustrates that Th22,Th17 cells may participate in the development of vitiligo,and are closely relative to its activity.Thus we can infer that the multiplies and activation of Th22 and Th17 may play an important role in the development of vitiligo.2.In vitiligo patients’ peripheral blood,Treg cells related factor expression and translation level is lower than healthy controls,and active patients is lower than stationary patients which demonstrate Treg cells may has influence on the development of the development of vitiligo,and are negative to its active.Thus infer that Treg cells and its related factors may decline in vitiligo patients’ peripheral blood and the function may be impaired,so that it can’t effectively restrain Th22,Th17,and other related amplification of helper T cells’ multiplies and activation,and participate in the development of vitiligo.3.Compared with healthy controls,in ordinary and segmental vitiligo patients’ peripheral blood,there were immune related factors associated with T cell have changes,and trend of changes are same.But ordinary vitiligo has a bigger change than segmental vitiligo.Thus suggests that T cell-mediated immune response play a role in development of two type of vitiligo: in t ordinary vitiligo,immune response may play a major role;For segmental vitiligo,other factors may be relatively more important than immune.4.In vitiligo patients’ peripheral blood,Th22,Th17 cells related cytokines levels are higher than the healthy controls,Treg cells related cytokines Treg cells is lower than the healthy controls,it shows that there are correlations between the three types of cells:Th22 cells and Th17 cells have dirert proportional correlation,both of them are inversely proportional to Treg cells.These three cells through mutual promotion and restrain,jointly participate in the pathogenesis of vitiligo.
