Objective1.Building EMs rat model of haemorrheological nature and blood stasis type and discussing on effects of EMs on endometrial capacity of model rats.2.Observing the effects of the TONGYUSHU on the window pinocytosis and LIF of the endometrium of EMs rat with haemorrheological nature and blood stasis type, comparing the differences and investigating the action mechanism of TONGYUSHU on rat endometrium receptive, and offering the reference for the clinical treatment.3.Observing the effects of the preventive medication of TONGYUSHU on on the window pinocytosis and LIF of the endometrium of EMs rat with haemorrheological nature and blood stasis type, and discussing the significance of early intervention. Methods1.40 SD female rats were randomly divided into blank control group, which only had 5 rats. Built the haemorrheological nature and blood stasis type EMs rat model, setting up in 35 in total, through autologous transplantation of endometrial and ice water soaking. At 3 days,5 days,4 weeks,6 weeks,8 weeks, randomly selecting 5 rats and dynamicly observing models and the pathological ectopic lesions.2.150 SD female rats were randomly divided into blank control group, which only had 18 rats. Built the haemorrheological nature and blood stasis type EMs rat model, setting up in 132 in total, through autologous transplantation of endometrial and ice water soaking.108 rats only after laparotomy were randomly divided into the model group for 4 weeks, the model group for 8 weeks, the low dose traditional Chinese medicine group, the middle dose traditionalChinese medicine group, the high dose traditional Chinese medicine group and the western medicine group. After 4 weeks of operation, the model group for 4 weeks was put to death at fourth days of pregnancy. The rest of the traditional Chinese medicine group were given 4 weeks intragastric administration of the setting dose, then were put to death at fourth days of pregnancy. Then take the left side of the uterus of rats, observing the pinocytosis vesicles expression by electron microscope and detecting LIF by immunohistochemical determination and the pathological ectopic endometrium. Western medicine group stopped medication for a week, then the pregnancy, sampling and testing as the former.3.90 SD female rats were randomly divided into blank control group, which only had 18 rats. The same method to build the haemorrheological nature and blood stasis type EMs rat model, setting up in 72 in total. Laparotomy exploration on 5 days after operation, choosing 18 rats which grow successfully as prevention group. They were given 4 weeks intragastric administration of the best dose and an ice water soaking after surgery a week. The rest 4 weeks after laparotomy exploration choosing 36 rats,18 of which were the model group for 4 weeks and were put to death at fourth days of pregnancy, sampling and testing as the former. The remaining 18 rats as high dose group, after 4 weeks intragastric administration, the pregnancy, sampling and testing as the former.Results1.The average success rate can reach 80% through autologous transplantation of endometrial and ice water soaking to build the haemorrheological nature and blood stasis type EMs rat model.2.In Ems rats model, the window period pinocytosis vesicles expression and the expression of LIF of the model rats were significantly lower than normal rats(P<0.05). After treatment, compared with the model group for 8 weeks, the traditional Chinese medicine groups pinocytosis vesicles mature cases increased and the high dose traditional Chinese medicine group were more obvious(P<0.05), The mature vesicle distribution area of the traditional Chinese medicine groups were also increased, the high dose group performance was more dense. The expression of LIF of the traditional Chinese medicine groups were rising, the high dose group effect was best, western medicine group was not obvious.3.After the treatment, the symptoms of the traditional Chinese medicine groups obviously relieved, but western medicine group was not obvious and the weight gained. The lesions were all narrowed down. Pathological observation showed the growth of ectopic endometrium restrained, the effects of the Chinese medicine high dose group and western medicine were best.4.After preventive treatment, compared with model group,, the window period pinocytosis vesicles mature cases of the treatment group and prevention group rats under electron microscope were increased(P< 0.05), the difference was statistically significant, but the prevention group occupied larger area and denser in the distribution of the pinocytotic vesicles; compared with the model group, the LIF expression of treatment group and prevention group were increased, prevention group was better than the middle and the Chinese medicine high dose group, the difference was statistically significant.5.Prevention group rats did not appear obvious symptoms of haemorrheological nature blood stasis, the lesions of the prevention group and the high dose group were reduced, Pathological observation showed the growth of ectopic endometrium restrained, the effect of the prevention group was better than the high dose group.Conclusion1.The espression of the pinocytosis vesicles and LIF in window period of endometrial tissue of the EMs rat with haemorrheological nature and blood stasis type are lower than the normal rats.2.Therapeutic medications of the TONGYUSHU can improve the pinocytosis vesicles and LIF in window period of endometrial tissue of the EMs rat with haemorrheological nature and blood stasis type.3.Preventive medications of the TONGYUSHU can more effectively improve the expression of the pinocytosis vesicles and LIF in window period of endometrial tissue of the EMs rat with haemorrheological nature and blood stasis type than the therapeutic medications.4.The TONGYUSHU Medicines can improve endometrial receptive and increase the conception rate by improving the expression of the pinocytosis vesicles and LIF of the model rats.
