ObjectiveThe purpose of research is to explore the effect of Fufangbaoshen particles (FFBS) on C-BSA heterogeneous serum immune of complex nephritic rabbits and Adriamycin nephritic rats by observing its’ Urinary protein, Serum creatinine,BUN and the histology of kidney, the level of associated protein factors to reveal the mechanism of its prevention and treatment of CGN.MethodsFirstly, Continuously injected C-BSA on the rabbits’ear vein for 6 weeks to make rabbit get nephritis, then divided into 6 groups:normal control group, model group, dexamethasone group,FFBS particles 1.2g/kg,0.6g/kg,0.3g/kg group. After 10 weeks gavage administration, then tested urine protein, Scr, BUN and kidney pathological changes, tested the expression of ICAM-1 and IL-6 from rabbits’ kidney tissue. Effects of FFBS particles was observed on C-BSA heterogeneous serum immune of complex nephritic in rabbits.Secondly, injected doxorubicin hydrochloride 5mg/kg on the rats’ tail vein once,to make rats get nephritis two weeks later, then divided into 6 groups:normal control group, model group, prednisone group,FFBS particles 1.5g/kg,0.75g/kg,0.375g/kg group. After 4 weeks gavage administration, then tested urine protein, Scr, BUN and kidney pathological changes, tested the expression of TGF-β1 and Nephrin from rats’kidney tissue. Effects of FFBS particles was observed on Adriamycin nephritic in ratsResultsFirstly, FFBS particles 1.2g/kg dose group could significantly reduced urine protein from C-BSA nephritic in rabbits(p<0.05 or p<0.01),. FFBS particles 1.2g/kg & 0.6g/kg dose group rabbits’Scr significantly reduce(P<0.01). FFBS particles 1.2g/kg dose group also could significantly reduced BUN(p<0.05).Secondly, FFBS particles 1.2g/kg dose group could significantly improve pathological changes from C-BSA nephritic in rabbits such as tubular dilatation, tubular atrophy, tubular epithelial cell degeneration and necrosis, renal tubule tubular, renal interstitial hemorrhage& congestion, renal interstitial fibrosis, swelling, Inflammation has been significantly improved(p<0.05). MASSON and SPA staining showed that Renal interstitial fibrous connective tissue proliferation, glomerular basement membrane thickening, glomerular solid plug, blocking, renal capsule expansion, tubular dilatation and edema also has been significantly improved.Thirdly,FFBS particles 1.2g/kg dose group could significantly reduced MID of ICAM-1 and IL-6 from C-BSA nephritic in rabbits(p<0.05).Fourthly, FFBS particles 1.5g/kg and 0.75g/kg dose group could reduced urine protein from Adriamycin nephritic in rats, Among this 1.5g/kg dose group with significantly reduction after the lst,2nd,3rd week of treatment (p<0.05),0.75g/kg dose group with significantly reduction after the lst,2nd week of treatment. FFBS particles 1.5g/kg dose group rats’ Scr significantly reduce(p<0.01). FFBS particles 1.5g/kg and 0.75g/kg dose group also could significantly reduced BUN(p<0.01).Fifthly, rats kidney HE staining:1.5g/kg dose group(2/9),0.75g/kg dose group(4/10), 0.375g/kg dose group(2/10), Most tubule cell swelling, obviously vacuolar degeneration Sixthly, Clearly visible within the bureaucratic protein casts. Each group remaining kidney tissue intact renal capsule.1.5g/kg dose group mesangial proliferation was not obvious, 0.375g/kg dose group part of mesangial proliferated mildly. Other animals tubular cell swelling degeneration,intraluminal protein casts,interstitial inflammatory cell infiltration: 1.5g/kg<0.75g/kg<0.375g/kg. FFBS particles 1.5g/kg dose group MASSON staining showed that Renal interstitial fibrous connective tissue proliferation also has been significantly improved with a lower MID.Finally, FFBS particles 1.5g/kg dose group could significantly promoted MID of Nephrin from Adriamycin nephritic in rats(p<0.05). FFBS particles 1.5g/kg and 0.75g/kg dose group could significantly reduced MID of TGF-β1 from Adriamycin nephritic in rats(p<0.05).ConclusionC-BSA heterogeneous serum immune of complex nephritic rabbits and Adriamycin nephritic rats can be considered as an animal model for human CGN. FFBS particles has a good preventive effect for this two kinds of mode,which could possessed a preventive effect for Urinary protein,Scr,BUN and the histology of kidney,its’Mechanismis possibly through intervening the expression of IL-6,ICAM-1, Nephrin and TGF-β1 this four protein factors closely associated with kidney disease.
