| ObjectiveThe present study is to evaluate the effects ofShen-yan-xiao-bai (SYXB) granules on nephropathy using RT-PCRand investigate the potential anti-nephropathy mechanismsabout podocyte-associated molecules.MethodsRats were injected with Adriamycin (6.5mg/kg) via the tail veinto induce nephropathy. Adriamycin induced nephropathic ratswere divided into model group, Benazepril group (0.9mg/kg),SYXB granules low dose group (1.8g/kg), SYXB granules high dosegroup (3.6g/kg).Body weight and urinary albumin/urinarycreatinine were measured every week. Selected three rats fromeach group who would be put to death in the3th and5th weekend,cutted the kidney for two pieces along the center line of thevertical, taked the right kidney in order to be detected of theqRT-PCR, while the other one to do the immunohistochemical.ResultsThe Shen-yan-xiao-bai (SYXB) granules can improve the usualmode and the weight of rats; SYXB granules can improve thepathology of kidney; Urinary albumin/urinary creatinine of thetreated groups in7weeks was less than the model group (P<0.05),the SYXB high dose group was better than the Lotensin group (P<0.05); In immunohistochemical, the Nephrin proteinexpression was higher than the model group in kidney (P<0.05),the SYXB high dose group was better than the Lotensin group(P<0.05); The Nephrin mRNA expression was higher than the modelgroup in kidney (P<0.05), the SYXB high dose group was betterthan the Lotensin group by RT-PCR; SYXB granules has less harmto the kidney than the drugs.ConclusionsSYXB granules can decrease the urinary albumin ofAdriamycin–induced nephropathy in rats; SYXB granules canprotect the podocyte by increasing the expression of nephrinmRNA and protein; The effectiveness of SYXB granules was betterthan the Lotensin, and the effectiveness of SYXB granules inhigh dose was better than the SYXB granules in low dose. |
