The Experimental Study On The Molecular Mechanism Of Eight Are Scattered Decoction On Intervention Drift Of Th1/Th2 Of Male SD Rats With Chronic Abacterial Prostatitis

Objective:Through the experimental study on the molecular mechanism of Eighte are Scattered Decoction on intervenetion drift of Th1/Th2 in male SD rats with chro-nic abacterial prostatitis, evaluatethe intervention effect, and explore the mechanism of Eight are Scattered Decoction in treatment of chronic abacterial prostatitis, so as to look for experimental and theoretical basis of Eight are Scattered Decoction in the treatment of chronic abacterial prostatitis.Method:15month- old male SD 60 rats, were randomly divided into blank group, model group, Qian LieTongYu capsule group, Eight are Scattered Decoction in low、 medium、high dose treatment groups, each group of 10 only. Except the blank group, the rest of the group rats was given with intraperitoneal injection of anesthesia and castration operation under sterile conditions, continued to give subcutaneous injection of estradiol benzoate 0.25mg/kg the day after the operation, once a day for 30 days.Last time of subcutaneous injection of estradiol benzoate ended and gavage began next day, blank group and model group were intragastrically infused with a saline content equaled to the Chinese Medicine content of experiment group; control group:each rat was given by intragastric administration of Qian Lie Tong Yu capsule suspension 3.9g/kg-d,equaled to 20 times of the amount of the adult weight 70Kg; Eight are Scattered Decoction low、medium、high dose group:each rat was given by intragastric administration of Eight are Scattered Decoction suspension3.12g/kg·d、 6.24g/kg·d、12.48g/kg·d,equaled to 5 times、10 times、20 times of the amount of the adult weight 70Kg.All groups were intragastrical orally once for 28 days. Continuous intragastric 28d,24h after the last administration,said the weight of each rat, then put it to death,removed the prostate tissue and said its wet weight.The prostate tissue was fixed by 4%　formalin,the pathological changes were observed under light microscope, IFN-γ、IL-4 content with Elisa method.Result:Compared with the blank group, the ratio of IFN-γ/IL-4 of the model group were significantly increased (P<0.05); compared with the model group, the control group,the ratio of IFN-y/IL-4 of decreased significantly (P<0.01), Eight are Scattered Decoction in the low dose group, the ratio of IFN-y/IL-4 did not change significantly (P>0.05), Eight are Scattered Decoction in the medium、high dose group, the ratio of IFN-y/IL-4 decreased significantly (P<0.01); compared with the control group, Eight are scattered Decoction in the medium、high dose group,the ratio of IFN-y/IL-4 did not change significantly(P> 0.05).Conclusion:Eight are Scattered Decoction in the appropriate dose can reduce the response level of Thl cells in the body,improve the body’s Th2 cells response level, adjusting the Thl/Th2 cells balance, so as to achieve the objective of treatment on chronic abacterial prostatitis.
