| Chinese medicine has the characteristics of multiple components, multiple ways, multiple targets and multiple effects, which makes the study on the substance basis of Chinese compound prescription, pharmacokinetics, biological effects and their relationship is quite complex. In view of these characteristics, how to establish the component index system and effect system of traditional Chinese medicine? How to integrate multiple index, multiple effect for PK-PD study? How to choose the appropriate data processing method and establish the PK-PD binding model adapt to the characteristics of the active ingredient group and network target pharmacodynamics? These are the problems that a traditional Chinese medicine multi-component PK-PD binding model needs to solve.Related research issues are complex and difficult, this paper creativity selected one of them to find the PD-PK markers through PK-PD model. According to the reasonable selection problem of Pharmacopoeia standards, Shuanghuanglian oral liquid is checked as the research object with a broad spectrum of antibacterial, antiviral and antioxidant effect. On the foundation of early literature research, consider abandon phillyrin, and select baicalin, forsythiaside A and chlorogenic acid as objects of study. Winnonlin software is used to analyze the pharmacokinetic parameters of three target components, and the routine PK-PD data analysis method, the partial least squares (PLS) method, genetic algorithm are comprehensive used. Through experiment and analysis, forsythiaside A is preliminary judged as antiviral PK marker, baicalin is judged as antioxidant PK marker.This paper creatively put forward reasonable choice issues according to the Pharmacopoeia standards, and creatively proposed the PK markers related to the effects. In the data processing, three different data processing methods were compared and analyzed, reasonable method selection for data provides a new angle of view. |
