This object was supported by Jiangsu Universities’ Research Findings Industrialization Promoting Projects "GAP study of Herba Taching and Herba Lophatheri" (No.JHZD09-11 to Q.N.W.) and the outstanding scientific and technologcial innovation team of resoures chemical researches on traditional Chinese medicine of universities and colleges in Jiangsu (2011). Herba Lophatheri is the dried stem and leaf of Lophatherum gracile Brongn., the family of Gramineae. According to Chinese medicinal theory, Herba Lophatheri affects the body by clearing away heat and reducing fire. It also has a relaxing effect and acts as a diuretic, and was often used to cure the fever, urodynia and buccal ulceration in clinical. Herba Lophatheri was one of the Edible Chinese herbal announced by the Ministry of Health, not only contained the necessary nutrients of food, but also the active constituents and bias of Chinese herbal. Herba Lophatheri was safe and reliable, which has obvious efficacy. But the method of quality evaluation is still single, the effective ingredient is not very clear, and pharmacological effects is not deeply. This paper carried out the research work are as follows:1. The resource survey was conducted on the main producing area of the herba, and briefly summarized. Having studied the available research, it finds the Ganzhou in Jiangxi Province、Chenzhou in Hunan Province and Tongren in Guizhou Province were the main areas where the commercial Herba Lophatheri in the current market came from. The other products were produced and marketed all by themselves. The system survey included annual output, recovery processing, store, commodity specification and price, using, survival condition, the problems of existence, and so on.2. Seven flavonoid glycosides were isolated and obtain, there were swertisin, swertiajaponin, tricin luteolin, luteolin-7-O-β-D-glucoside, vitexin, isovitexin, while swertisin was obtained from Herba Lophatheri first.3. More comprehensive accurate HPLC finger-print was established, cluster analysis and factor analysis was conducted.4. A method for simultaneous quantitative determination of seven polyphenol constituents from Herba Lophatheri by HPLC was developed.5. Gray relation analysis was used to explor the relationship of the content of secondary metabolites and ecological factor. The data about longitude and latitude, above sea level and leading climate factor was collected to proceed the quality evaluation and gray relation analysis.6. IR fingerprint of Herba Lophatheri was firstly built. A dual-index sequential analysis method for the common and variant peak ratios, with the infrared spectrums of different samples as a standard was established with the second order derivative spectrum, to classify the Herba Lophatheri from different areas into four groups, distinguish it with its counterfeit drug, then different Herba Lophatheri extract from bamboo leaf extract. Otherwise, a method for the determination of leaf and stem of Herba Lophatheri by near infrared (NIR) spectroscopy is presented, which preliminary explored the holistic differences about the compositiones of the leaves and the stems.7. Research on the flavonoids dynamic accumulation law of Herba Lophatheri. Simultaneous determinate the seven polyphenols in different growth periods samples for analysis. The results showed that the content of the herb were on the rise.8. This study also contained a part of pharmacology experiments. The results showed that total flavone extracted from Herba Lophatheri can enhance the contractility of isolated notmal rat hearts, and significantly increased LVSP,+dp/dtmax,-dp/dtmax index, but swertiajaponin can not. This exlporation offers a new concept to the pharmacological and pharmacodynamics research about Herba Lophatheri total flavone.9. Research on the protective effects of Herba Lophatheri total flavone on myocardial ischemia-reperfusion injury in rats. The myocardial ischemia-reperfusion injury model in rat were established by ligating left anterior descending (LDA) coronary artery. The activities of lactate dehydrogenase (LDH), creatine phosphokinase (CK), superoxide dismutase (SOD), maleic dialdehyde (MDA), glutathione peroxidase (GSH-PX) and nitric oxide (NO). Total flavone of Herba Lophatheri can inhibit CK and LDH leaks out in myocardial, reduce LDH, CK activity and MDA content in serum and myocardial tissue, and improve SOD, GSH-PX and NO concentration. Total flavone of Herba Lophatheri exerts significant cardioprotective effects against acute ischemia myocardial injury in rats.
