Influence Of Herb-cake-separated Moxibustion On The Immunologic Function In Immunosuppressive Rabbits

Object: In the experiment, herbal cakes made of Drugs of nourishing kidney Yin were put on the points in the form of moxibustion, such as Shenque, Guan Yuan, Zusanli,etc, to observe the changes of peripheral blood cells series influenced by cyclophosphamide; the changes of IgG、IgM, the complement C3、C4 in serum immunoglobulin, and the changes of spleen morphometry. To study the mechanism of immune function improved by herbal cake separated moxibustion.Methods: According to the random number table, 50 white rabbits were randomly divided into 5 groups. They were Blank Group, Immunosuppressed Model Group, Herbal cake-separated moxibustion Group, Moxibustion Group and Sham cake-separated moxibustion Group. Each group except the blank group was given intraperitoneal injection ofcyclophosphamide(Cy) 60mg/kg every morning, for seven consecutive days. Two days after the completion of modeling, three groups were treated every other day by herbal cake-separated moxibustion, moxibustion, sham cake-separated moxibustion treatment, a total of 10 times except the Immunosuppressed Model Group. When treatment were finishied, collected Abdominal venous blood and removed spleens after animal anesthesia the next day. To observe the changes of peripheral blood cells series influenced by cyclophosphamide; the changes of IgG、IgM the complement C3、C4 in serum immunoglobulin and the changes of spleen morphometry.Results:1. After making model, the indexs of WBC, NEU%, RBC, HGB, HCT of each group was significantly lower than the Blank Group(P < 0.01), after moxibustion with medicinal cake, it could significantly rebound the content of WBC, NEU%, RBC, HGB, HCT of immunosuppressive blood cells series of immunosuppressed rabbits, compared with the Blank Group, there was no significant difference; the RBC of immunosuppressed rabbits increased significantly after moxibustion, but compared with the Blank Group, there was still significant difference; Fake cake separated moxibustion Group and Immunosuppressed Model Group had no significant difference, but,the difference was significant compared with the Blank Group.2. After making model, the levels of IgG, IgM, C3 were significantly lower than that of Blank Group(P ﹤ 0.01), it could obviously improve the level of IgG ofimmunosuppressed rabbits after moxibustion with medicinal cake on the points,such as Shenque, Guan Yuan, Zusanli,Pishu,Shenshu, and the difference was obvious compared with the Blank Group(P﹤0.01), the difference of indexs of IgM, C3 was not obvious; the levels of IgG, IgM, C3 of immunosuppressed rabbits after moxibustion were significantly higher(P < 0.01),but IgG and IgM had no obvious difference compared with the Blank Group, and C3 had significant statistical significance(P < 0.01); The C3 of immunosuppressed rabbits had rebounded.3. The results of spleen form metrology showed that the average size of white pulp and the radius of splenic corpuscle were significantly reduced in the Model Group, lymphatic cells of lymphatic sheath around the arteries were also significantly reduced compared with the Blank Group, a lot of hemosiderin particles in red pulp(old hemorrhage), part of the red pulp had tracts of massive intramedullary hemorrhage. After moxibustion with medicinal cake, the average size of white pulp had a significant increase, lymphatic cells of lymphatic sheath around the arteries were significantly increased and were dense, there was no obvious difference compared with the Blank Group, a significant increase in the radius of splenic nodule, the amount of hemosiderin particles in red pulp decreaseed, intramedullary hemorrhage of red pulp decreased significantly; After moxibustion, splenic corpuscle of white pulp had a significant increase in the average size and radius, there was significant difference compared with the Blank Group, the difference of the count of lymphatic cells of lymphatic sheath around the arteries had no sense of statistics; After fake cake moxibustion, the average area of white pulp and the radius of splenic corpuscle expanded, but compared with the Blank Group, the difference was statistically significant, lymphatic cells of lymphatic sheath around the arteries were significantly increased, and had no obvious difference compared with the Blank Group, large amounts of hemosiderin particles were observed in the red pulp and large amount of intramedullary hemorrhage.Conclusions:1. Moxibustion with medicinal cakes could improve the level of WBC、NEU%、RBC、HGB、HCT of blood cell series of immunosuppressed rabbits, improve the level of IgG、IgM、C3 of Humoral immunity, the average size of white pulp,the radius of splenic corpuscle, and the amount of lymphatic sheath cells around the central arteriole of spleen, made all the indicators above return to normal, to improve the immune function.2. Moxibustion could obviously rebound the level of RBC、IgG 、IgM、C3 of immunosuppressed rabbits, the average size of white pulp,the radius of splenic corpuscle,and could improve the immune function.3.Compared with the Moxibustion Group,the Moxibustion Group with medicinal cake had more obvious effect on improving the level of WBC、NEU%、RBC、HGB、HCT of of blood cell series of immunosuppressed rabbits, the level of C3 of Humoral immunity, and the amount of lymphatic sheath cells around the central arteriole of spleen.
