Objective:To investigate the influence of silodosin on the pharmacokinetics of levofloxacin in treatment of rat’s chronic bacterial prostatitis (CBP).Methods:A total of ninety-six SD rats were used to develop the models of chronic bacterial prostatitis and then they were divided into two groups randomly:control group (only treated with levofloxacin, n=48) and experimental group (treated with both silodosin and levofloxacin, n=48). Six rats from each group were enthanized to collect their prostate samples to make tissue homogenate at each time point:0.125,0.25,0.5,1,2,4,8 and 12h after levofloxacin administration. High performance liquid chromatography (HPLC) were applied to detect the concentrations of levofloxacin and 3p97 software program was used to calculate the pharmacokinetic parameters.Results:The levofloxacin concentration-time curves in rats’ prostate of the two groups were both fit the one-compartment model. The major pharmacokinetic parameters of the control group and experimental group as follows:half-time (t1/2)were (4.34±0.25) h and (5.02±0.28) h, time to peak (tpeak) were (0.73±0.20) h and (0.86±0.05) h, clearance rates (CL) were (1.74±0.19) mg/h and (0.64±0.06) mg/h, maximum concentration (Cmax) were (3.62±0.3) mg/kg and (8.47±0.51) mg/kg, area under the curves (AUCo~12) were (21.58±2.00) mg·h/kg and (56.63±4.30) mg·h/kg, respectively. Compared with the control group, silodosin in the experimental group prolonged the t1/2 (P<0.05) and obviously increased the Cmax, decreased the CL of levofloxacin in prostate tissues (P<0.01). However, there were no significant differences (P>0.05) between the two groups in the tpeak.Conclusion:Silodosin can significantly elevate the concentration of levofloxacin in the prostatic tissues of rats with CBP.
