Effects Of TCF21 On Invasion And Metastasis Of Human Hepatocelular Mammary Carcinoma Cell Line SMMC-7721 Cells And Its Mechanism

OBJECTIVE:Hepatocellular carcinoma (HCC) is a common malignant worldwide tumour of the high mortality. Transcription factors 21 (TCF21), which is located in 6q23.2, is a newly discovered tumor suppressor genes in recent years. It was found methylated and deactivated in a wide variety of tumor tissues. It has been reported that TCF21 was high methylated and down-regulated expression in HCC tissue, but high expression in normal liver tissue relatively. Our research is to analyze the impact of TCF21 gene and its related mechanism on proliferation, migration and apoptosis in hepatocellular carcinoma cell line SMMC-7721 cell.METHODS:We stability transfected pcDNA3.1 (+) TCF21 and pcDNA3.1 (+) plasmid into SMMC-7721 cells by using Iipofectamine2000 and respectively named them TCF21 over expression group, empty vector group, untreated group. Expression of TCF21 and mRNA and protein were investgated by RT-PCR and Western blot. Cell proliferation ability was detected by MTT assay. Wound Healing was used to test cell migration ability, flow cytometry was applied to observe cell apoptosis rate. Western blot was used to examine expression of related proteins.RESULTS:TCF21 present high expression in both gene and protein level after TCF21 transfection in SMMC-7721 cells. According to the results of MTT assay, the proliferation ability of over expression group was relatively less than control group. Wound Healing results has shown that TCF21 can significantly inhibit tumor cell migration ability (P< 0.01). Compared with empty vector group and untreated group by Flow cytometry, the apoptosis rate of over expression group was statistically significant which was 50.08±1.13 (P< 0.05). Western blot results demonstrated that TCF21 can raise the expressions of KISS 1, p53, and down-regulate MMP-9 (P< 0.05).CONCLUSION:Tumor-suppressor genes TCF21 could inhibit the proliferation, migration of SMMC-7721 hepatocellular carcinoma cells and promote its apoptosis.
