The Effects Of Mir-5481on The Proliferation,Apoptosis And Migration Of Highly Metastatic Lung Cancer Cell Line95D

Objective:To investigate the effect of has-miR-5481(miR-5481) on the proliferation, apoptosis and migration of human highly metastatic95D lung cancer cells.Methods:miR-5481expression vector plasmid was transiently transfected into highly metastatic95D lung cancer cells by lipofectamine2000in vitro. The expression of miR-5481in95D lung cancer cells was detected by RT-PCR after transfection.Low metastatic95C lung cancer cells,95D lung cancer cells and95D lung cancer cells transfected by miR-5481nagetive control plasmid as control groups, the change on the proliferation capacity of95D lung cancer cells was determined by the method of MTT. The apoptosis of95D lung cancer cells was accessed by the flow cytometry. And the migration capacity of95D lung cancer cells was analyzed by Trans well assay.Results:1. The expression level of miR-5481in95D group was defined as l,the expression of miR-5481in NC group and miR-5481transfected group was (3.032±0.198) and (2418.539±171.096)Compared with95D group and NC group,the expression of miR-5481markedly increased in miR-5481transfected group (P<0.05)2. MTT test showed the proliferation of95D lung cancer cells had no statistical difference miR-5481transfected group and NC group (P>0.05).The result suggested miR-5481had no dramatical effect on the proliferation of95D lung cancer cells.3. The flow cytometry indicated that the apoptosis of cells in95C group,95D group,95D group and miR-5481transfected group was (20.75±9.96)%,(10.33±0.25)%,(39.27±1.65)%and (53.77±1.89)%.Compared with95C group,95D group and NC group,the apoptosis of cells highly increased in miR-5481transfected group (P<0.05).The study indicated miR-5481can significantly promote the apoptosis of95D lung cancer cells.4. Transwell test demonstrated the migration of cells in95C group,95D group, NC group and miR-5481transfected group was (32.50±0.71),(43.50±2.12),(43.05±1.34) and (18.35±2.33), compared with95C group,95D group and NC group, the migration of cells in miR-5481transfected group extremely decreased (P<0.01).The result showed miR-5481can observably inhabit the migration of95D lung cancer cells.Conclusion:The overexpression of miR-5481can promote the apoptosis of95D lung cancer cells and suppress its migration.
