Objective: Through the analysis of the RT-PCR(real time quantitative polymerase chain reaction) detection’s positive rate and sensitivity of HCMV-DNA in saliva, provide the basis for the detection of neonatal congenital CMV infection and select sensitive samples to assist in diagnosis of neonatal congenital CMV infection.Method: 1.Using synchronous blind method, 108 neonates’ s blood, urine, saliva samples were collected in Neonatal intensive care united of Fujian Provincial Maternity and Child Care Center during 2014.05.01~2015.03.01;2. Through 6 kinds of methods in detection of HCMV(RT-PCR text sample in saliva, urine and blood、chemiluminescence method to detect serum HCMV-Ig M and HCMV-Ig G、Check fresh urine exfoliated cell to find the characteristic inclusion body of HCMV),calculated the positive rate of each method. 3. according to the final diagnosis cases of neonatal congenital CMV infection, calculated the sensitivity of each method.Result: The newborns in experimental group(27 cases) were diagnosised congenital cytomegalovirus infection, the control group(81 cases) were negative by 6 kinds of methods in detection of HCMV. The results of detection:①the 108 cases use RT-PCR text sample in saliva, urine and blood HCMV-DNA for detection of HCMV positive rate was 13.9%(15/108)、12.0%(13/108)、7.4%(8/108),detection sensitivity was 55.6 %(15 / 27) 、 46.1 %(13 / 27) 、 29.6 %(8 / 27); ② The 108 cases use chemiluminescence method to detect serum HCMV-Ig M and HCMV-Ig G for detection of HCMV positive rate was 6.5 %(7/108) 、 2.8 %(3/108), detection sensitivity was 25.9 %(7 / 27)、 11.1 %(3 / 27);③ the 108 cases with HCMV inclusion in fresh morning urine method for detection of HCMV positive rate was 1.9%(2/108),detection sensitivity was7.4%(2/27).Conclusion: 1.the positive rate and sensitivity of RT-PCR for detection of HCMV-DNA in saliva is greater than other 5 kinds of detection methods;2.RT-PCRmethod in the detection of neonatal congenital CMV infection specimens choose,saliva should be preferred,followed by urine,then blood;3.If the condition is allowed, the multiple samples and methods should be detected at the same time to improve the detection rate of HCMV and decrease the false negative.
