Objective: To explore the relationship between susceptibility to lung cancer and methylation of p16, RASSF1 A genes in Chinese population.To study the extent of influence of passive smoking to the methylation of p16 and RASSF1 A genes of mice. Methods:(1) Meta-analysis method was used to study the relationship between susceptibility to lung cancer and methylation of p16, RASSF1 A gene in Chinese population.(2) Pyrosequencing assay method was used to detect the extent of influence of passive smoking to methylation of p16 and RASSF1 A gene in mouse lung tissue. Results:(1) Result of Meta-analysis: 10 studies met our inclusion criteria, 549 NSCLC cases and 513 controls about p16 methylation were brought into the study. Odds ratios(ORs) of p16 gene methylation were obtained from case and control groups was 6.76, 95%CI(4.79%~9.54%), statistical analysis showed significant difference between the two groups(p﹤0.05).(2) 8 studies met our inclusion criteria, 777 NSCLC cases and 596 controls about RASSF1 A gene methylation were brought into the study. Odds ratios(ORs) of RASSF1 A gene methylation were obtained from case group and control group was 4.46, 95%CI(3.35%~5.94%), statistical analysis showed significant difference between the two groups(p﹤0.05).(3) p16 gene methylation test results as follow: the methylation of p16 gene was not detected in blank control group, low dose group, middle dose group and high dose group.(4) RASSF1 A gene methylation test results as follow: the methylation of RASSF1 A gene was not detected in blank control group, low dose group, middle dose group and high dose group. Conclusion: There was closely relationship between methylation of p16, RASSF1 A gene and incidence, development of lung cancer in Chinese population. The influence of passive smoking on methylation of p16, RASSF1 A gene in mouse was not obvious.
