Objective:Observation of temperature-sensitive ion channel TRPV3(thermosensitive ion channnel TRPV3) in the human knee joint cartilage and synovial tissues, TRPV3 expression meaning in human cartilage and synovial tissues were discussed.Methods:Collect different ages normal articular cartilage tissue 10 cases of synovial tis sue five cases, Knee cartilage tissue five cases and five cases of synovial tissue o f patients with osteoarthritis, the cartilage tissue specimens were in each case in t he medial tibial plateau sampling, synovial tissue specimens were sampled in the joint capsule layer, put in the freezer-70℃ save. Each specimens are used Safran in "O" staining verify degenerative changes. First, selected the five cases of OA c artilage tissue as osteoarthritis group, 60-69 year-old group five cases selected as normal control group, Using RT-PCR, Western blot detection TRPV3 expression i n normal and OA cartilage and synovial tissue, immunefluorescence and immunoh-istochemistry was to detected the expression of TRPV3 channel in normal cartila ge and OA cartilage, and then detected by Western blot TRPV3 expression differe nces between the two groups.Then the samples were normal human cartilage tissu e were analysis by Western blot, and the resulting bands were gray value analys is, the no statistically significant difference in the gray values are grouped, a total of two groups of five cases, respectively: 30-39 age group, 60-69 age group, an d finally detected by Western blot TRPV3 in normal human cartilage tissue age t-rend.Results:Under the microscope show Safranin "O" staining osteoarthritis group articular cartilage is not normal,dyeing and level uneven, poorly among cellular staining was significantly lighter, glycosaminoglycans reduced. Staining in normal control group of normal cartilage tissue was uniformly red, pale staining shallow, longitudinal section stained enhanced deepest deep stain, showed no changes of degenerative; RT-PCR,Western blot, immunofluorescence and immunohistochemical methods in cartilage and synovial tissues were detected TRPV3 channel proteins, immunofluorescence staining showed TRPV3 uniformly distributed on the cell membrane; the coomassie brilliant blue staining control the use of the test sample volume, TRPV3 strip drawn each case based on the gray value for statistical analysis showed that the expression of osteoarthritis group TRPV3 is higher than the normal control group, t test was statistically significant(P <0.05). TRPV3 expression were detected two ages normal cartilage tissue samples showed that with age, TRPV3 expression showed a growing trend, t test was statistically significant(p <0.05).Conclusions:TRPV3 and osteoarthritis are closely linked,it may play a role in the development of osteoarthritis which provide new targets for articular cartilage injury prevention and treatment. |
