Objective:Pancreatic tumors,especially pancreatic cancer,have insidious onset,rapid progression and high degree of malignancy,which seriously threaten the life and health of patients and bring a huge economic burden to the society.The purpose of this study was to explore the expression of TRPV3 in pancreatic tumors,and to clarify the role of TRPV3 channel protein in the biological behavior of pancreatic cancer cells,so as to provide a new direction for the diagnosis and treatment of pancreatic cancerMethods:The pancreatic tumor pathological samples which recorded in the pathology department of the First Hospital of Shanxi Medical University were collected,and the clinical data of related patients also be focused,The TRPV3 expression in pancreatic tumors include pancreatic cancer,Solid pseudopapilloma of the pancreas and Negative marginal tissue were studied with immunohistochemical method,using SPSS statistical software to analyze the relationship between TRPV3 expression in different tissue and the clinical pathological features of patientsFurther cell experiments were performed,cultivate pancreatic cancer cells and normal pancreas cells,using Western blot and qt-polymerase chain reaction(PCR)to detect TRPV3 expression in these cell lines,select the appropriate pancreatic cancer cells,and transfected TRPV3 interference sequence and compared with the control group,then performed the different cell function experiment respectively,in order to make clear TRPV3 role in pancreatic cancer cell biology behavior mechanismResults:A total of 49 pathological samples were used for immunohistochemical experiments,among which 20 were pancreatic cancer tissues,with positive TRPV3 expression accounting for 75.0%(15/20),and 15 were solid pseudopapilloma tumor of the pancreas,with positive TRPV3 expression accounting for 53.3%(8/14).The positive expression of TRPV3 accounted for 35.7%(5/14)of the 15 patients with normal pancreas(paracancer negative margin).The expression of TRPV3 in pancreatic cancer and pancreatic solid pseudopapilloma was not statistically significant(P=0.181),the expression difference between pancreatic cancer and normal pancreas was statistically significant(P=0.022),and the expression difference between solid pseudopapilloma and normal pancreas was not statistically significant(P=0.340).20 cases related patient's clinical pathologic data were collected,the statistical analysis suggests that the relationship between TRPV3 expression in pancreatic cancer and the patient's age,gender,the discretion of CA19-9,tumor size,lymph node metastasis,nerve infiltration and vascular invasion,tumor clinical stage and degree of differentiation in clinical pathological features were no significant statistical correlationAfter conventional cell culture,Western blot and qt-pcr detection showed that TRPV3 expression in pancreatic cancer cells was higher than that in normal pancreatic cells,and TRPV3 expression levels at mRNA and protein levels were highest in PANC-1 cells.After transfected PANC-1 into NC-siRNA and TRPV3-siRNA successfully,the results of cellular functional experiments showed that the proliferation were enhanced,apoptosis,invasion and migration of PANC-1 cells were weakened after TRPV3 knockdown,and there were no significant changes in cell cycleConclusion:The expression level of TRPV3 in different pancreatic tissues was different,and the expression level of TRPV3 in pancreatic cancer was higher than that in normal tissues(paracancer negative margin),and the difference was statistically significant.TRPV3 expression levels of mRNA and protein in pancreatic cancer were higher than that of normal pancreatic cells.Combined with the above results,the high expression of TRPV3 channel protein in pancreatic cancer tissues and cells may be one of the potential causes of pancreatic cancer.Through various cellular functional experiments,it was found that TRPV3 had little effect on cell cycle.Although TRPV3 could inhibit the proliferation of pancreatic cancer cells,it could significantly enhance the invasion and migration of cancer cells and reduce the apoptosis of cancer cells.Combined with the above results,it is suggested that TRPV3 channel protein may promote the development of pancreatic cancer rather than inhibit it,which may lead to highly malignant biological behavior of pancreatic cancer.
