Objective:People found that clinical hypothyroidism during pregnancy is associated with a variety of obstetric complications, suc h as gestational diabetes, gestational hypertension disease, placental abruption, spontaneous abortion, premature birth, low birth weight, etc. With gestational hypertension disease is a peculiar disease during pregnancy, and may be accompanied by organs such as heart, liver, kidney, brain damage, is the leading cause of maternal and perinatal deaths, but its pathogenesis is not clear, but there are many scholars confirm the placenta cytokine secretion imbalance may be one of the cause of gestational hypertension disease, Show the placenta Th1 cytokines such as TNF-α, IL- 2 secretion increased, while the Th2 cytokines secreted decrease such as IL- 10. Related, the results indicate the specific construction form of hypothy-roidism effect on pregnancy outcome are diverse, mechanism is not clear, may partly associated with thyroid hormone directly involved in placenta. This research mainly by immunohisto chemical method of determination of rat placenta and thyroid tumor necrosis factor alpha(TNF-α) and interleukin 10(IL- 10) to explore the relation between the two diseases. Methods:Using propylthiouracil tablet to establish models of hypothyroidism during pregnancy rats, by using the immunohisto chemical SABC method detected the expression of TNF-α and IL- 10 of placenta and thyroid tissue in 24 cases of normal pregnant rats, hypothyroidism pregnant rats, treatment of hypothyroidism pregnant rats, merger pih hypothyroidism pregnant rat. Results:1. The normal group, the hypothyroidism group, hypothyroidism treatment group, hypothyroidism merger PIH group,TNF-α and IL- 10 of placenta and thyroid diverse by Kruskal Wallis rank and inspection, P < 0.05, the difference is statistically significant, TNF-α, IL- 10 level of four groups of samples in placenta and thyroid is not the same;2. To compare TNF-α、IL – 10 of hypothyroidism group and normal group in placenta and thyroid respectively, P < 0.05, the difference is statistically significant, hypothyroidism group of TNF alpha levels in placenta and thyroid higher than normal group, and IL- 10 level was lower than those of normal group, to compare the hypothy-roidism treatment group and normal group TNF-α,IL10 in placenta, P > 0.05, the difference is not significant; comparison in thyroid, P < 0.05, the difference is significant;3. Compared TNF-α, IL – 10 of hypothyroidism group with the group of hypothy-roidism treatment in placenta, P < 0.05, the difference is statistically significant, Com-pared in thyroid, P > 0.05, the difference is s not significant, hypothyroidism group have higher levels of TNF-α than treatment group, and IL- 10 level is low, compared TNF-α, IL – 10 of the hypothyroidism group with the hypothyroidism group merged with PIH in placenta and thyroid, P > 0.05, the difference is not significant.Conclusions:1. When the pregnancy with hypothyroidism cytokine secretion imbalance may be causes of pregnancy- induced hypertension disease and other adverse pregnancy outcomes.2. Thyroid hormone may affect the secretion of cytokines, placenta directly involved in the development of the placenta.
