Objective To investigate the effects of treadmill training at different dose on neurological function and expression of kalirin-7 after cerebral ischemia-reperfusion in rats, to identify the possible mechanism underpinning the neural benefits of intensive training in rats after cerebral ischemia-reperfusion.Methods To establish animal model of cerebral ischemia-reperfusion, middle cerebral artery occlusion(MCAO) using intraluminal thread was performed on the left side in rats. 45 male MCAO model rats were randomly divided into 3 groups, including MCAO model group, common training group and intensive training group(15 rats in each group).Another 15 rats subjected to sham operation were assigned to be in sham group. Each of the 4 groups was randomly divided into 3 subgroups, including3 d, 7d and 14 d subgroups. MCAO model group and sham group did no motor training; common training group did treadmill training once daily; intensive training group did treadmill training twice a day. Neurological assessment was performed using Zausinger scores; Expressions of kalirin-7 and kalirin-7 m RNA were detected by western blot and RT-PCR.Results Zausinger scores of each motor training group were higher than those of MCAO model groups at the same time points(P <0.05);At 3th day, there was no statistical significant between intensive training group and common training group in Zausinger scores; the expression of kalirin-7 and kalirin-7m RNA among motor training groups and MCAO model group had no statistical significant.At 7th day, Zausinger scores of intensive training group were significantly higher than those of common training group(P < 0.05); the expression of kalirin-7 of each motor training group was significantly higher than that of MCAO model group(P < 0.05),and intensive training group was significantly higher than common training group(P < 0.05); the expression of kalirin-7m RNA of intensive training group was significantly higher than that of common training group(P<0.01).At 14 th day, Zausinger scores of intensive training group were significantly higher than that of common training group; The expression of kalirin-7 and kalirin-7m RNA of each motor training group was significantly higher than that of MCAO model group(P < 0.05), and intensive training group was significantly higher than common training group(P<0.05).Conclusion 1.Motor training can improve the injured neurological function of rats after middle cerebral artery occlusion(MCAO); Of all the training groups, intensive training group obtained the best effects. 2. Motor training can increase the expression of kalirin-7 and kalirin-7m RNA, which point out that motor training can affect the reshaping process of brain by increasing synapse formation,thus can improve the injured neurological function; intensive training group obtained the best effects.
