Objective To observe the effects of intensive training on the degree of depression and expressions of kalirin-7 in hippocampus after post stroke depression in rats, and to investigate its possible mechanism.Methods Forty male Wistar rats were randomly divided into a post-stroke depression(PSD)group, a routine training group, an intensive training group and a sham-operation group(n=10 for each group). Rats ischemic model were established middle cerebral artery occlusion by the intraluminal thread method except the sham-operation group. The PSD model was established by applying chronic mild stress to the individually housed rats. The PSD group and the sham-operation group did not do any training, the routine training group did training once a day, the intensive training group did training twice a day. On first day, 14 th day, and 28 th day, ethology test was used to evaluate the degree of depression after PSD model; kalirin-7 in the hippocampus were detected by Western blot.Results After 14 d training, compared with the sham-operation group, there were statistically significant differences for the results of ethology test of the other groups(P<0.05). Compared with the PSD group, there were statistically significant differences for the results of OFT(55.71±3.94)s and FST(55.71±3.94)s of the routine training group(P<0.05), compared with the PSD group and the routine training group, there were statistically significant differences for the results of ethology test of the intensive training group(P<0.05). After 28 d training, compared with the sham-operation group, there were statistically significant differences for the results of ethology test of the PSD group and the routine training group(P<0.05). Compared with the PSD group, there were statistically significant differences for the results of ethology test of the routine training group and the intensive training group(P<0.05), compared with the routine training group,there were statistically significant differences for the results of ethology test of the intensive training group(P<0.05). In addition, on 14 th and 28 th day, the expression of kalirin-7 of the the routine training group and intensive training group were significantly higher than that of the PSD group(P<0.05), the expression of intensive training group were higher than that of the routine training group(P<0.05).Conclusion The intensive training can improve the degree of depression of the PSD rats,and increase the expression of the kalirin-7 in hippocampus.
