| Abstract:Objective:To investigate the relation between DcR3 and mechanism of tumorigenesis and development of salivary gland tumors and its clinicopathological significance, and to provide guidance for clinical diagnosis, treatment and prognosis of salivary gland tumors by examining expression of DcR3 protein and mRNA in salivary gland tumors.Methods:SP immunohistochemical method and in situ hybridization method were used to investigate the expression of DcR3 protein and mRNA in 51 cases of salivary gland tumors and 5 cases of normal salivary gland tissues,and we analyzed relationship of between expression of,DcR3 protein and mRNA and clinicopathological parameters of salivary gland tumors.Results:â‘ DcR3 protein positive staining was yellow or brown granules,located in the cytoplasm, and were expressed mainly in the cytoplasm of epithelial tumor cells in salivary gland tumors and the cytoplasm of epithelial cells of the Leap tube and corrugated tube of normal salivary gland tissue.â‘¡DcR3mRNA positive hybridization signal was yellow or brown granules,mainly located in the cytoplasm, the nucleus can be seen in a small amount of expression, and were expressed mainly in the cytoplasm of epithelial tumor cells in salivary gland tumors and the cytoplasm of epithelial cells of the Leap tube and corrugated tube of normal salivary gland tissue;â‘¢The positive rate of DcR3 protein in malignant salivary gland tumors was higher than that in benign salivary gland tumors (χ2=4.647,P=0.031 <0.05),and expression of DcR3 protein in malignant salivary gland tumors was higher than that in benign salivary gland tumors(Z=-2.505, P=0.012<0.05).â‘£Expression of DcR3mRNA in malignant salivary gland tumors was higher than that in benign salivary gland tumors(Z=-2.207,P=0.027<0.05).Expression of DcR3 protein and mRNA was not related to their type of organization in Warthin adenoma,basal cell adenoma and pleomorphic adenoma(x2=0.387,P=0.824>0.05.χ2=1.325, P=0.516>0.05).⑦Expression of DcR3 protein and mRNA was not related to their type of organization in mucoepidermoid carcinoma and adenoid cystic carcinoma(Z=0.868,P=0.385>0.05.Z=-0.600,P=0.549> 0.05).â‘§Expression of DcR3 protein and mRNA was related to malignant degree in malignant salivary gland tumors,the expression in high-grade malignant group was higher than that in low-grade malignant group(Z=-2.577,P=0.010<0.05.Z=-2.677,P=0.007<0.05);⑨Expression of DcR3 protein and mRNA was related to clinical stage and lymph node metastasis in malignant salivary gland tumors:Expression of DcR3 protein and mRNA in clinical stageâ…¢-â…£was higher than that in clinical stageâ… -â…¡(Z=2.294,P=0.022<0.05.Z=-2.470,P=0.014<0.05),and expression of DcR3 protein and mRNA with lymph node metastasis group was higher than that without lymph node metastasis group(Z=2.242,P=0.025<0.05.Z=-2.486,P=0.013<0.05),but expression of expression of DcR3 protein and DcR3mRNA was not related to sex, age of patients with salivary gland tumor and size,location of tumors (P>0.05).â‘©There was a significant positive correlation between the expression of DcR3 and expression of DcR3mRNA in salivary gland tumors(rs=0.573,P=0.000<0.01).Conclusions:â‘ DcR3 protein and DcR3mRNA were mainly expressed in the cytoplasm of epithelial tumor cells in salivary gland tumors and the cytoplasm of epithelial cells of the Leap tube and corrugated tube of normal salivary gland tissue.â‘¡Expression of DcR3 protein and mRNA in malignant salivary gland tumors was higher than that in benign salivary gland tumors,and expression of high-grade malignant group was higher than that of low-grade malignant group in malignant salivary gland tumors.So we speculated that DcR3 may be involved in tumorigenesis and development of a salivary gland tumors and malignant transformation of salivary gland tissue cells.â‘¢Expression of DcR3 protein and mRNA was related to clinical stage and lymph node metastasis in malignant salivary gland tumors,but not related to sex, age of patients with salivary gland tumors and size, location of tumors.So we speculated that DcR3 may be a potential reference to judge the prognosis of patient gland tumors and to predict metastatic potential of cells in malignant salivary gland tumors. (4)Expression of DcR3 protein was significantly consistent with that of DcR3mRNA in salivary gland tumors,and they both can be valuable detected markers in salivary gland tumors. |
