Objective:To investigate An Shen capsule’s effects on Schizophrenic s’ cognitive function Methods:Using randomized, trial study method with which 68 cases of patients with Schizophrenics were randomly divided into experiment group(n= 34 cases) and control group(n=34 cases). All patients were given a 12-week risperidone treatment( 2mg oral risperidone once, and twice a day), except for the same basic risperidone treatment, experiment group was also given Anshen capsule 4 pills(1.6g) oral Take one three times a day. Before and after 12 weeks treatment, the clinical effects and cognitive function were evaluated with Positive and Negative Syndrome Scale(PANSS), Wisconsin Card Sorting Test(WCST) Wechsler Memory Scale-III(WMS-III), Number cancellation test(NCT), and also the adverse drug reaction and safety index of Anshen capsule were measured with Treatment Emergent Symptom Scale(TESS). Results: 1 34 patients were studied in the experiment group of which 2 patients dropped the test. 34 patients were brought into control group, of which 4 patients gave up.The difference at the age, education level, the disease course and other conditions between the two groups is too little to have statistical significance(P >0.05), and the comparability of the two groups was good enough before the treatment. 2 Compare the evaluate results of executive function(WCST) before and after the 12-weeks treatment within the group: the experiment group had some improvement on Total Number of Correct, correcting number classification, continuous number errors, Failure to Maintain Set showing on the baseline before the treatment, this difference(improvement) was statistically significant(P <0.05); The control group also had some improvement on correcting number, correcting number classification, continuous number errors, Failure to Maintain Set,in which all the difference(improvement) was is statistically significant(P <0.05) except for the Failure to Maintain Set(P >0.05). Comparison of the improvement on executive function index between the two groups after 12 weeks treatment: all the difference(improvement) was statistically significant(P <0.01) except for the correcting number classification(P >0.05) among the two groups. Therefore, it proved that the experiment group had been improved more than the control group. 3 Compare the evaluate results of attentional function(NCT) before and after the twelve-week treatment within the group: the net scores of the two groups raised in different levels, and the the difference(improvement) was statistically significant(P <0.05); The error rate declined in different levels, but only the difference(improvement) in the experiment group was statistically significant(P <0.01); Comparison of the improvement on attentional function index between the two groups after 12 weeks’ treatment: the difference of the net scores and the error rate of the two groups was statistically significant(P <0.01),Therefore, it proved that the net scores and the error rate of the experiment group had been improved more than the control group’s. 4 Compare the evaluate results of memory function(WMS-Ⅲ) before and after the twelve-week treatment within the group: both of the Anterograde score and Retrograde score increased in different levels together with the total scores, and the difference(improvement) has statistical significance(P<0.05); Comparison of the improvement on memory function index between the two groups after 12 weeks’ treatment: the difference(improvement) of the Anterograde score of the two groups were statistically significant(P<0.01),but the difference of the Retrograde score of the two groups were not statistically significant(P>0.05), Therefore, it proved that the Anterograde score and the total score of the experiment group improved more than the control group, while the difference(improvement) of the Anterograde score is in the same level as it is of the control group. 5 Compare the evaluate results of the curative effect on mental symptom(PANSS) before and after the 12 weeks’ treatment within the group: both of the Positive score and Negative score have some statistically significant decrease together with the Pathological score and the total score(P<0.01),in the experiment group, and so is it for the control group(P<0.05).The difference of the positive score of the two groups before and after 12 weeks’ treatment is not statistically meaningful(P>0.05),while the difference of the negative score and Pathological score together with the total score is statistically significant. Comparing the decreasing ratio of the PANSS score of the two groups after the treatment, the effective ratio in the experiment group is 90.65%, and the control group is 70%, so it obviously proved that the effective rate of the experiment group is higher than the control group with significant difference(P <0.01). 6 Assess the drug safety with TESS: there is no serious side effects during the treatment, only 4 cases occured side effects in the experiment group with the side-effects incidence of 12.5%, while 14 cases occurred in the control group with the incidence of 46.7%, the difference of the two groups is statistically significant(P <0.01). Conclusion: 1 Using Risperidone only or together with An Shen capsule both can improve Schizophrenics’ cognitive function, but using An Shen capsule together is more efficient. 2 Using Risperidone only or together with An Shen capsule both can improve Schizophrenic’psychiatric symptom, but using An Shen capsule together is more efficacious. 3 An Shen capsule as adjunctive therapy to Risperidone is safe in clinical medication.
