Objective:To investigate the expression and clinical significance of NGAL gene in the tissue of squamous cell carcinoma and adenocarcinoma,and preliminary verify its value as a lung cancer indications matter in determine prognosis. Methods:1. Using the quantitative PCR to detect the adenocarcinoma, lung squamous cell carcinoma on lung and analyzed the expression of NGAL gene.2. Using immunohistochemical technique to detect the corresponding expression of NGAL gene in 55 cases of lung squamous cell carcinoma, 63 cases of lung cancer and 118 cases of cancer tissue and adjacent,and found the relationship between non-small cell lung cancer and its different pathological types expression in tumors contact with clinical data.3. Using ELISA(enzyme-linked immunosorbent assay) to measured 98 cases of non-small cell lung cancer patients with peripheral blood NGAL levels, and compared 80 cases of healthy subjects. Analysis of changes in lung cancer patients NGAL serum and its correlation with clinical parameters, the initial judgment of the diagnostic value of serum NGAL. Results:1.The curves of NGAL gene amplification smooth in quantitative PCR experiments are typically seen in the early logarithmic amplification, logarithmic amplification phase and plateau phase, NGAL expression in non-small cell lung cancer tissues was significantly higher than that next to the lung tissue, an average of 7.386 ± 4.150 fold difference was statistically significant(t = 4.680, P<0.05).2.In immunohistochemistry experiment we found the lung squamous cell carcinoma positive expression rate is 54.54%(30/55), adjacent control the expression rate of only 3.63%(2/55), The positive rate of 77.77%(49/63) in lung adenocarcinoma, next to its cancer control tissue expression rate of only 4.76%(3/63). With lymph node metastasis in non-small cell lung cancer of NGAL expression was 84.1%(58/69), and in the absence of lymph node metastasis, the positive rate was 53.0%(36/49), the difference was significant(P<0.05); NGAL in Ⅲ stage(TNM staging) non-small cell lung cancer was 93.0%(40/43) was significantly higher than the I / Ⅱ stage(TNM staging) non-small cell lung carcinoma which rate was 58.6%(44/75)(P<0.05) 3. ELISA test showed that the NGAL expression in lung cancer group were significantly higher than the control group, NGAL expression and patient age and sex, but not with lung cancer specific pathological type, TNM stage and lymph node metastasis are closely related, after surgery compared to NGAL levels before surgery, there are significant differences. Conclusion: 1. There have present NGAL gene expression in Squamous cell carcinoma and adenocarcinoma of lung in RNA and protein levels. 2.There is a big difference in the expression of NGAL gene expression in adenocarcinoma and squamous carcinoma of the lung adenocarcinoma and adenocarcinoma significantly higher than squamous cell carcinoma. 3. Squamous cell carcinoma and peripheral serum NGAL expression levels of patients before surgery was significantly higher than normal, and reduced quickly after operative, Based on these findings, the expression of NGAL may be an important tumor marker to predict NSCLC.
