Objective: To investigate the genetic polymorphism of 24 STR loci on chromosome Y of She and Han population in Fujian and explore its application in paternity testing.Methods:(1) DNA of 228 male individuals of She population and 211 male individuals of Han population with unrelated bloodlines was extracted and amplified by 24 Y-STR loci multiplex PCR system. Then PCR products were separated and genotyped with capillary clectrophoresis and fluorescence detection technology.(2)Genetic indexes of 24 STR loci of She and Han population such as AF, GD and HD were calculated, which differences with that of other populations were analyzed by reference to domestic and foreign literatures.(3) 5 cases with difficultly judged paternity relation were analyzed by 24 Y-STR detection system mentioned above.Results:(1) In Fujian She population, the allele frequency of the 24 Y-STR loci was 0.0044~0.7193, and the gene diversity was 0.4539~0.9181. In Fujian Han population, the allele frequency of the 24 Y-STR loci was 0.0047~0.9573, and the gene diversity was 0.5218~0.9612.(2) 24 Y- STR loci of 228 male individuals of She population in Fujian were observed and 177 haplotypes were found, with 1 time of 148 haplotypes, 2 times of 20 haplotypes, 3 times of 3 haplotypes, 4 times of 2 haplotypes, 5 times of 1 haplotype, and 6 times of 3 haplotypes. Accordingly, indexes of haplotype diversity and discriminative power were deduced to be 0.9964 and 0.7763 respectively. 24 Y- STR loci of 211 male individuals of Han population in Fujian were observed and 211 haplotypes were found. Accordingly, indexes of haplotype diversity and discriminative power were 1 and 1.0000 respectively. Interestingly, 1 haplotype appeared in both She and Han population.(3) The allele frequency distribution of 24 Y-STR loci of Fujian She population was compared with that of Han population and other domestic and foreign populations by 104 times comparison, of which 98 times with significant differences(P <0.05), 6 times with no significant dfferences(P >0.05). The allele frequency distribution of 24 Y-STR loci of Fujian Han population was compared with that of She population and other domestic and foreign populations by 104 times comparison, of which 92 times with significant differences(P <0.05), 12 times with no significant dfferences(P >0.05).(4) Through the detection of 24Y-STR system, 5 cases with difficult paternity judgement were indentified successfully, of which 1 case and 3 cases were confirmed with no paternity relation and paternity relation respectively, and the other 1 case was judged to be the same paternal.Conclusions:(1) There are relatively high genetic polymorphism and some differences of the allele frequency and gene diversity of 24 STR loci on chromosome Y of She and Han population in Fujian, which might contribute to promoting the study of the genetics in Fujian She and Han population.(2) Haplotype distribution of 24 STR loci on chromosome Y in Fujian Han population is better than that of She population.(3) 24 Y-STR loci detection system has higher cumulative probability of paternity exclusion, which application could improve the detection efficiency of paternity testing. Accordingly, Y-STR loci might be used as a valuable supplement to autosomal STR loci in difficult paternity judgement and paternal family survey.
