Objective To investigate the distribution and relationship of SNP locus on interleukin-1 a(IL-1a),interleukin-1?(IL-1?),interleukin-6(IL-6),interleukin-10(IL-10),?-tumor necrosis factor(TNF-?)and rheumatoid arthritis(RA).Methods(1)Selecting 155 RA patients(RA group)and 170 healthy subjects(control group)in Han population of Quanzhou,Fujian Province.Detecting IL-10 rs1800893?IL-6 rs1800796?IL-6 rs2066992?IL-1?rs16944?IL-1?rs1143623?IL-1a rs17561?IL-1a rs1894399 ? IL-1a rs3783550 ? IL-1a rs1800587 ? TNF-?rs2009658 ?TNF-?rs1041981 with whole blood tri-primer allele-specific amplification(TRIP-ASPCR).(2)Hardy-Weinberg genetic balance test was used to analyze the data of 11 SNP loci in 325 subjects by chi-square test,t-test and logistic regression with SPSS19.0 software and network biological information tool to analyze the distribution of genotype and allele in the Han population in Quanzhou,Fujian Province.The correlation index(OR)of the incidence of RA was statistically analyzed with the statistical significance of P <0.05.P>0.05 was not significantly correlated.(3)Link disequilibrium analysis and haplotype analysis were performed with SHEsis software.The linkage disequilibrium coefficient was significant with D'= D / Dmax,D'> 0.70,r2> 0.33 was significant,and haplotype analysis was OR > 1 is positively correlated with RA.Results(1)The SNPs of IL-6 rs1800796,IL-1a rs3783550,IL-1a rs1800587 and TNF-?rs1041981 in the control group were in accordance with the Hardy-Weinberg equilibrium test.IL-10 rs1800893 and IL-1a rs1800587 meet the(P> 0.05)Hardy-Weinberg equilibrium test.(2)Allele frequencies of IL-10 rs1800893,IL-6 rs1800796,IL-6 rs2066992,IL-1?rs16944,IL-1a rs17561,IL-1a rs3783550,TNF-?rs2009658 and TNF-?rs1041981 were significant differente between the control group and the RA group(P <0.05).There was no significant difference in frequency between the control group and the RA group of IL-1?rs1143623?IL-1a rs1894399 and IL-1a rs1800587(P> 0.05),and the correlation with RA was not obvious.(3)There was linkage disequilibrium between IL-6 rs1800796 and IL-6 rs2066992(D '= 0.820,r2 = 0.151).There was no linkage disequilibrium between IL-1?rs16944 and rs1143623(D' = 0.121,r2 = 0.009).There were no linkage disequilibrium between the four SNP of IL-1ars17561,IL-1a1894399,IL-1ars3783550 and IL-1ars1800587(D?=0.06,0.390,0.107,r2 =0.00,0.0.15,0.0001).There was no linkage disequilibrium between TNF-?rs2009658 and TNF-?rs1041981(D '= 0.465,r2 = 0.003).The distribution of haplotype frequency of IL-6 rs1800796 and IL-6 rs2066992 was statistically significant(P <0.05).GT was positively correlated with RA(P=3.33e-16,OR=22.765).The distribution of haplotype frequencies of IL-1a rs17561,IL-1a rs1894399,IL-1a rs3783550 and IL-1a rs1800587 was statistically significant in the control group and RA group(P <0.05).G T A A was positively correlated with RA(P = 0.0037,OR = 29.413).The haplotype frequency distribution of TNF-?rs2009658 and TNF-?rs1041981 was statistically significant in the control group and RA group(P <0.05).AC was positively correlated with RA(P = 0.041,OR=1.443).There was no significant difference in the distribution and haplotype frequencies of IL-1?rs16944 and IL-1?rs1143623 between the control group and the RA group(P> 0.05).Conclutions(1)IL-10 rs1800893,IL-6 IL-1 rs2066992,IL-1? rs16944,IL-1a rs17561,IL-1a rs3783550,TNF-? rs2009658 and TNF-? rs1041981 were associated with the incidence of RA in the Han population of Quanzhou,Fujian Province.allele G of IL-10 rs1800893,allele G of IL-1 rs1800796,allele T of IL-1a rs17561,allele G of IL-1?rs16944,allele G of IL-1a rs17561,allele C of IL-1a rs3783550,allele C of TNF-?rs2009658 and allele C of TNF-?rs1041981 can be used as potential genetic markers for the diagnosis of RA in Quanzhou,Fujian Province.(2)Rs17561,rs1894399,rs3783550 and rs1800587 of IL-1a,rs1800796 and rs2066992 of IL-6,rs2009658?rs1041981 of TNF-? and their joint-action were positively correlated in the incidence of RA of Han population,Quanzhou,Fujian Province.
