Objective: To investigate the genetic polymorphism and genetic analysis of12STR loci onchromosome X in Fujian population and evaluate the application value in paternity testing..Methods: Samples from200unrelated individuals (104males and96females)of Hanpopulation and100unrelated individuals(50males and50females) of She population wereextracted and amplified with12X-STR loci multiplex PCR system,PCR products wereseparated and genotyped with capillary clectrophoresis and fluorescence detection technology.Statistic the genetics index of Fujian Han population and She population,such as PD、PE、PIC,ect.(2) Father-daughter and mother-son(daughter) forensic paternity testing cases weredivided into three groups according to the appraisal autosomal STR classificationresults.Testing12X-STR loci in100cases which confirmed paternity relation and100caseswhich excluded paternity relation and50cases which unjudged paternity relation.It could beobserved whether the genotyers of12X-STR loci of parental generation were coinside withthe genotyers of descendant or not.RESULT:(1)In Fujian Han population,the polymorphism information content (PIC)of the12X-STR loci was0.48~0.91,the female discrimination power was0.6975~0.9896,the malediscrimination power was0.5274~0.9251. The expected probability of exclusion in trio cases(MECtrio) was0.4482~0.9203and0.3088~0.8557in duo cases.(2)In Fujian Shepopulation,the polymorphism information content (PIC) of the12X-STR loci was0.40-0.89,the female discrimination power(PDf)was0.6769~0.9853,the male discriminationpower (PDm) was0.5119~0.9101. The expected probability of exclusion in trio cases(MECtrio) was0.4272~0.9034and0.2911~0.8307in duo cases.(3)The genotyers of12 X-STR loci drew the same conclusion as the autosomal STR classification results in100“confirmed”and100”excluded”cases.There were5cases which could make a judgement ofexclusion in50cases which unjudged paternity relation.Conclusion:The12X-STR loci have high genetic polymorphism in Fujian Han populationand She population There are different genetic distributions and indexs among the12X-STRloci. they are valuable for the research of the paternity testing,evaluating the cumulativeprobability of paternity exclusion. The12X-STR loci are powerful and reliable for paternitytesting,especially for the testing of special paternity cases....
