Objective:1. To explore the effect of high blood pressure and electrocardiogram under the long-term exposure in low concentrations of arsenic.2.Analysis and comparison of the change of band-3 protein expression in the rat chronic arsenic exposure, looking for the chronic arsenic exposure early biomarkers;3. Study of chronic arsenic exposure on the molecular mechanism of rat hemoglobin damage. Found a new way of arsenic poison mechanism.4. To explore the effect of GSH on rats. Discusses the damage of red blood cells under the long-term exposure in low concentrations of arsenic.Methods:180 people were choose as exposed group and 194 as control group,all of which have been living there at least 25 years. The investigator are professional. Using electronic blood pressure meter and a 12-lead electrocardiogram meter, these person had a medical examination by clinical doctors trained professionally. Blood pressure and electrocardiogram(ECG) were both measured and analyzed by investigator. Then spss13.0was used on blood pressure and electrocardiogram analysis.Will be a healthy weight is 160g- 180 g of 40 SD rats of male and female half only in accordance with the method of randomized into control tranches, low dose tranches,middle dose tranches, high dose tranches of four tranches, each tranches in accordance with the method of free water virus infected dose of each group were 0μg/L,10μg/L,60μg/L, 360μg/L. Animals infected two months, blood collection and abdominal aorta.Analyzing and comparing different groups of rats weight general toxicity index; Using Sysmex Xt-2000 i automatic blood cell analyzer determination of different groups of rat red blood cell parameters: number of red blood cell(RBC), hemoglobin(HGB), RBC deposited(HCT), average red blood cell volume(MCV), mean red blood cell hemoglobin content(MCH), mean red blood cell hemoglobin concentration(MCHC), red blood cellwidth coefficient of variation(RDW); With reduced glutathione(GSH) kit by using enzyme-linked immunosorbent(Elisa) in the determination of plasma glutathione etc. The damage index of red blood cells; Adopt the method of western blot- analysis and comparison between groups of rats RBC band 3 protein and hemoglobin α1 chain(Hb A1)protein content.Results:1. The Abnormal blood pressure rate in the exposure group was 47.78% and it was37.63% in the control group. The difference between this two abnormal blood pressure rates has the statistically significant.(P<0.05);2. The difference between exposure group and control group on single abnormal rate of systolic blood pressure has the statistically significant(P<0.05), the difference between this two group on the single Diastolic abnormalities rate doesn’t has the statistical significance(P>0.05).3. The ECG abnormalities rate in the exposure group was 53.3% and it was 22.68% in the non-exposed Group. The difference between these two groups has the statistically significant(P<0.05).Compared with the control group, the Q-t lengthened in the ECG,Sinus arrhythmia, ST-T segment changes, premature and the abnormal rate of low voltage were increased significantly. And also it has the statistical significance.(P<0.05).4. The general toxicity in ratsThe infected group rats weight at the same time the difference had no statistical significance(P > 0.05); The infected group rats heart, liver, spleen, lung, kidney viscera coefficient difference is not significant(P > 0.05).5.Red blood cell parameters in ratsCompared with the normal control group, number of red blood cell(RBC), hemoglobin(HGB), RBC deposited(HCT), average red blood cell volume(MCV), mean red blood cell hemoglobin content(MCH), mean red blood cell hemoglobin concentration(MCHC), red blood cell width coefficient of variation(RDW).each group of infected rat red blood cell parameters has not yet occurred obviously abnormal change(P > 0.05).6.Red blood cell protein in ratsCompared with the control group, 360μg/L band- 3 content in arsenic infected groupwas obviously higher than that of control group, the difference was significant statistically(p < 0.05); 360μg/L arsenic content of Hb A1 infected group was obviously higher than that of control group and 10μg/L arsenic infected group, the difference was significant statistically(p < 0.05);7.GSH in ratsCan by measuring the GSH content in plasma of rats, according to the results of60μg/L, 360μg/L arsenic content of GSH infected group was obviously less than control group and 10μg/L arsenic infected group, the difference was statistically significant(p <0.05).Conclusions:1.The low exposure of arsenic can cause blood pressure and ECG abnormalities in the crowd. Low exposure of arsenic is closely associated with the occurrence of cardiovascular disease.2.The chronic arsenic exposure can have toxic effects on rat red blood cells, can cause erythrocyte band- 3 protein and Hb A1 protein content increase.3. The chronic arsenic exposure rat red blood cell oxidative damage occurs, GSH is a major reduction of red blood cells in the body material, according to the results of 60μg/L,360μg/L arsenic content of GSH infected group was obviously less than control group and10μg/L arsenic infected group, the difference was statistically significant(p < 0.05),research has shown that oxidative damage of red blood cells.
