Objective :Objective to investigate the expression of GPC3,、AFP in hepatocellular carcinoma、intrahepatic cholangiocarcinoma、cirrhosis, and to compare the differences of expression of GPC3,、AFP in hepatocellular carcinoma、Intrahepatic cholangiocarcinoma、cirrhosis.Method:Based on 50 cases of primary hepatocellular carcinoma, 20 cases of intrahepatic holangiocarcinoma, 22 cases of nodular cirrhosis, immunohistochemistry method for GPC3, AFP detection, the positive expression of GPC3 in liver cancer detection at ifferent stages, different degrees of differentiation, expression of GPC3 、 AFP in epatocellular carcinoma, intrahepatic cholangiocarcinoma and nodular cirrhosis.Results:The specific expression of GPC3 in hepatocellular carcinoma, the expression of sensitivity and specificity were 84%, 97.7%, compared with the expression of GPC3 in intrahepatic bile duct cell carcinoma, nodular cirrhosis, the difference was statistically significant(P<0.05). GPC3 protein expression in hepatocellular carcinoma associated with gender, age, liver function classification, cirrhosis, tumor size, tumor number,vascular invasion, TNM staging, HBs Ag positive aspects of the difference was not statistically significant(P>0.05). GPC3 in the very early and early, positive rate of interim and advanced hepatocellular carcinoma was 82.4%, 84.9% respectively, significantly higher than the expression of AFP protein, the difference was statistically significant(P<0.05). Combined detection of GPC3 and AFP can be diagnosed early and increases sensitivity of early, middle and advanced stage of hepatocellular carcinoma was88.2%, 87.8%. The positive rate of GPC3 in high differentiation, differentiation, poorly differentiated hepatocellular carcinoma were 75%, 85%, 81.8%, significantly higher than the expression of AFP protein, the difference was statistically significant(P<0.05).Combined detection of GPC3 and AFP can be differentiated, the positive rate of diagnosis sensitivity of differentiation, poorly differentiated hepatocellular carcinoma is up to 75%,90%, 90.9%. GPC3 intrahepatic cholangiocarcinoma compared each stage, GPC3 in the very early and early middle period, the positive rate of intrahepatic cholangiocarcinoma was 0%, 6.25% respectively, and the expression of AFP protein in intrahepatic cholangiocarcinoma, there was no significant difference(P>0.05). The positive rate of GPC3 in high differentiation, differentiation, poorly differentiated intrahepatic cholangiocarcinoma were 25%, 0%, 0%, and the expression of AFP protein in intrahepatic cholangiocarcinoma, there was no significant difference(P>0.05).Conclusion:The high expression of GPC3 in hepatocellular carcinoma tissues, there may be involved in the development of hepatocellular carcinoma.The high expression of GPC3 in hepatocellular carcinoma tissues, the important role in early diagnosis of hepatocellular carcinoma, to distinguish between benign and malignant early lesions important role.
