Objective:Research changes SOD and MDA pre and pro the fat-loading and its relationship with atherosclerosis.Methods:10 healthy male New Zealand white rabbits, weighing 3.0-3.5kg, high-fat( 1%cholesterol, 10% egg yolk, 89% of the diet) continuously feeding 12 weeks, and in the initial and the 4th、8th、12th week to give fat meal load(containing 2% cholesterol, 20%egg yolk, 78% basal diet). Initial experiments and 4 weeks after giving fat meal load fasting 8 hours at 8 weeks, 12 weeks, in the pre-load and 2、4、6、8th hour the levels of SOD and MDA measured by the marginal ear vein blood after load. The initial experiment and 4th、8th、12th week were detected by abdominal aorta(Chin scores), the animals were sacrificed isolated aortic end of the experiment(12thweek), HE staining vessels Surface Hardening situation.Results:1. In the process of hardening of the arteries, changes of plasma MDA pre and pro the fat-loadingWith the progress of arteriosclerosis, the level of MDA increases,the highest at the8 th week(P<0.05), suggesting that the progress of oxidative stress may be associated with arteriosclerosis.The level of MDA: on the initial and 12 th week pro the fat meal,the level of MDA at 2thhour increased(P<0.05), 4thto 6thhour decreased, the highest at the8 th hour; on the 4th and 8th week pro the fat meal, the level of MDA increased( P<0.05), the highest at 2th hour, and then decreased gradually; suggesting that early(2th hour) pro fat meal oxidative stress is stronger.2. In the process of hardening of the arteries, changes of plasma SOD pre and pro the fat-loadingWith the progress of arteriosclerosis, the level of SOD decreased,especially on the 8th week( P<0.05), and the level of SOD decreased,especially at the 2thand 4thweek(P<0.05),suggesting that the decrease of SOD may be associated with arteriosclerosis,pro the fat-loading, the level of SOD showed a further decline.3. In the process of hardening of the arteries, changes of the ratio of MDA/SOD pre and pro the fat-loadingDuring the experiment,the ratio of MDA/SOD increased, especially on the 8thweek( P<0.05).on the 4th week pro the fat-loading, the ratio of MDA/SOD increased especially at the 2thand 4thhour(P<0.05), on the 8thweek increased, especially at the 2th.suggesting that the MDA and SOD may be associated with progress of oxidative stress during arteriosclerosis. And the fat-loading can make the degree of stress increased,especially in the early time(2th and 4th hour).4.abdominal aortic ultrasoundDuring the experiment, the change of the abdominal aorta in experimental animals :from smooth lining becomes rough, IMT gradually thickened, experiment first eight weeks,12 thweek could be observed plaques, wall damage appears as a process of gradually increasing.5. The process of atherosclerosis before and after fat meal MDA load change and arteriosclerosis( ultrasound score) correlated(1)4thweek fasting and postprandial 8thhour negative correlation, The rest was a positive correlation(especially at the 4th hour)(2)8thweek was positively correlated with postprandial 4thhour, The rest was a negative correlation(especially at the 6th hour)(3)12thweek was no correlation between postprandial 6thhour, The rest was a negative correlation6. The process of atherosclerosis before and after fat meal SOD load change and arteriosclerosis( ultrasound score) correlated(1)4th week was positively correlated with postprandial 4,8th hour, the rest was a negative correlation(especially at the 6thhour)(2)8th week were highly negatively correlated with fasting postprandial(especially at the 8thhour)(3)12th week negatively correlated, postprandial 2th hour no correlation, The rest wasa positive correlation(especially at the 2thhour)7. The macroscopic observation of experimental animal’s abdominal aortaCompared with the initial and the end of the experiment(12th week), the abdominal aortic profile of experimental animals showed visible patches.8. abdominal aortic pathologyInitial : intimal normal endothelial cells continuously intact, and no space plasma phenomena and foam cells, normal arteries; 12 th week: wall injury is serious, significant intimal thickening, endothelial cells and smooth muscle cell proliferation, appeared " empty plasma phenomena, " shows a lot of foam cells.Conclusion:1. During the experiment,about experimental animals which during 8 weeks, the level of MDA showed a trend to increase and the level of SOD showed a trend to reduce; after 8weeks the level of SOD increased and the level of MDA reduced,suggesting that the level of MDA and SOD was in mutual balance.2. Pro the fat meal early may have a big influence on peroxidation of lipid. With the progress of arteriosclerosis, peroxidation can come up at 2thhour pro the fat-loading...
