With sufficient unsaturated fatty acids and SOD,animal liver which was a kind of rich nutrition food and a source of high quality full protein,has an extensive application value. This research was about the preparation technology of liver albumen meal which has high protein content and SOD activity.We also detected the active ingredients of the liver ethanol-extract and studied on effect of preparation process on fatty acid,in order to research and development the new function of animal liver.In this research,ethanol deposit method was used to fix the protein of liver.The single factor of experiment and orthogonal experiment was used to screen and optimize the ratio of solid to liquid,mix time,temperature,and ethanol concentration and so on.Finally,we drew the conclusion that the best process condition is the mix time:12h;the ethanol concentration:70%;the ratio of solid to liquid:1:5;under normal temperature.The concentrations of protein left with different ethanol concentration were qualitatively detected with SDS PAGE;the concentrations of SOD left were qualitatively detected with PAGE and NBT staining analysis.It showed that extractions from different ethanol concentration had the same protein concentration and the SOD activity.Then the deposition was freezing-drying and became the liver albumen meal.By PAGE and NBT staining,SOD kept activities after freeze-drying.It also indicated that activity of SOD from liver albumen meal could be kept at normal temperature and in the dark.The content of ingredient of liver albumen meal was detected by Kjeldahl nitrogen method,Soxhlet extraction method,SOD-kit method and direct dry method.The result showed:the preparation of protein was 53.57%,the crude lipid content was 9.05%,the total SOD activity was 119.41 U/mg,and the water content in liver albumen meal was 3.22%.Under the best process conditions,the main nutritional components of ethanol-extract have been researched initially.The GC(gas chromatography) detected the fatty acid in liver. It showed that chicken liver has many kinds of fatty acids in which some odd carbon fatty acids existed.The ratio of unsaturated fatty acids in chicken liver was 57.05%and higher than that of the saturated fatty acids.In contrast with chicken liver,chicken liver extracted by alcohol also has many kinds of fatty acids,and the ratio of unsaturated fatty acids in it was 41.96%which was lower than that of saturated fatty acids.By HPLC,fat-solvable and water-solvable vitamins in ethanol-extract of chicken's live were detected.It showed that the vitamin A content was 6.03 IU/g,the vitamin D content was 6.38 IU/g and the nicotinic acid content was 1.04μg/g in ethanol-extract of chicken's liver.
