Objective:With the help of the wound tissue of rats model, to observe the experimental results of morphology and histology, and through immunohistochemical method to detect the expression of CD68 and VEGF, and by RT-PCR technique to detect the expression level of CD68, CD34 + and VEGF and α-SM actin after 5 days. Studying ginseng and astragalus healing promoted soup in the role of rat skin incision healing process, discusses the mechanism ginseng and astragalus healing promoted soup of the wound tissue repair. To promote the ginseng and astragalus soup provides the theory basis for further clinical promotion, and provide new train of thought for the further development of traditional Chinese medicine.Methods:96 rats were randomly divided into 6 groups: blank group, model group, ginseng and astragalus healing promoted soup the high, medium and low dose group and vitamin C group with 16 rats in each group. After the success of the model building, cage and medicine feeding respectively. After the treatment of 3 days, 5 days, 7 days and 10 days, randomly select 4 rats. Observing the condition of incision healing and getting materials. By HE and Masson,Sirius red and toluidine blue staining for histologic observation; By immunohistochemical method to detect CD68, the expression of VEGF; By RT-PCR technique test 5 postoperative CD68, CD34+, VEGF and α-SM actin expression level. All measurement data results in `x±s, using SPSS 17.0 statistical software for data processing. If the result satisfy normal distribution and f test, using the single factor analysis of variance. If not satisfied, and the rank and inspection, P<0.05 for the difference is statistically significant. Results:Ginseng and Astragalus healing promoted soup the decoction high dose group, middle dose group and vitamin C group, incision scar healing time is shorter, the scar of the touch is relatively soft, good elasticity, low dose group of scar is relatively rigid, poor elasticity, but is still better than the model group; The 3nd, 5th, 7th and 10 th day postoperatively, the ginseng and astragalus healing promoted soup the decoction of each dose group, and vitamin C group have better CD68, VEGF expression than the model group, and the difference has statistical significance(P<0.05). Ginseng and Astragalus healing promoted soup the decoction high dose group has better CD68, VEGF expression than the middle dose group; The ginseng and Astragalus healing promoted soup the decoction middle dose group has better CD68, VEGF expression than the low dose group. And the difference has statistical significance(P<0.05). The ginseng and astragalus high dose group and vitamin C group has no significant difference in CD68, VEGF expression(P>0.05). The 5th day postoperatively, the ginseng and astragalus healing promoted soup the decoction of each dose group, and vitamin C group have better CD68、CD34+、VEGF andα-SM actin expression than the model group, and the difference has statistical significance(P<0.05). Ginseng and Astragalus healing promoted soup the decoction high dose group has better CD68、CD34+、VEGF andα-SM actin expression than the middle dose group; The ginseng and Astragalus healing promoted soup the decoction middle dose group has better CD68、CD34+、VEGF andα-SM actin expression than the low dose group. And the difference has statistical significance(P<0.05). The ginseng and astragalus high dose group and vitamin C group has no significant difference in CD68、CD34+、VEGF andα-SM actin expression(P>0.05).Conclusion:Ginseng and Astragalus healing promoted soup can effectively shorten the healing time of skin incision. This could accelerate its inflammation, increasing the activity of macrophages, stimulating the incision local blood vessels, fibroblasts and interstitial collagen formation. Ginseng and astragalus healing promoted soupg can improve skin incision healing in the process of acceleratin, and it has certain concentration-response relationship. The soup is better than vitamin C in promoting and accelerating scar tissue inflammation.
