Objective: To observe the effects of Zhenqifuzheng capsule(ZQFZ) on blood routine、 cognitive ability and central neurotransmitters 5-HT、Ach in chronic Benzene poisoning mice and to explore its possible mechanisms for regulating cognitive ability.Methods: 8-12 weeks age,male and healthy Kunming mice Seventy were by random number table randomly divided into 5 groups, i.e. the normal control group, the model group, the Western medicine joint ZQFZ treatment group,the ZQFZ treatment group,the Western medicine treatment group, 14 in each group. The mice were exposed to manually simulated high concentrations of benzene(benzene concentration=2000mg/m3)for eight hours every day, twenty-one successive days, the mouse that were selected the WBC drop to 6×109/L below, PLT drop to 200×109/L below,HBG drop to 98g/L below were the chronic benzene intoxication mice model. After the success of the molding, according to routine blood and behavioural indicators, the scores of similar mice were by random number table randomly divided into the normal control group, the model group, the Western medicine joint ZQFZ treatment group,the ZQFZ treatment group, as well as the western medicine group 5 groups, each group of 10.Mice in ZQFZ treatment group administered with ZQFZ soup 0.068g/g.d respectively by gastrogavage.Equivalent to clinical adult dosage of 9.1times.Mice in the Western medicine treatment group were administered with 0.08mg/g.d leucogen,0.2mg/g.d batyl alcohol and 0.08mg/g.d vitamin B4 by gastrogavage. Equivalent to clinical adult dosage of 9.1times.Mice in the Western medicine joint ZQFZ treatment group were administered with ZQFZ, leucogen, batyl alcohol and vitamin B4.Mice in the normal control group and the model group were feed normally.Each group is continuous treated for three weeks.To prevent the spontaneous recovery afte stop benzene poisoning,the rest four groups all continue to feel in high benzene environment during the treatment except the normal control group.Respectively after successful modeling and drug intervention, To detect cognitive function in mice by open field test and Morris water maze test. Brain tissue of mice in TPH2 m RNA, Ach E m RNA expression detected by Q-PCR method. Test the Hippocampal neuronal morphology,finally.Results:(1)Comparison of the blood routine of mice in each group: After quiet room inhaled benzene exposure three weeks,Compared with the normal control group, the WBC,PLT,HBG obviously decreased in the model group(P<0.05). After drug intervention three weeks, Comparison with the model group, the WBC,PLT,HBG were increased in all treatment group(P<0.05),Compared with the ZQFZ treatment and the western medicine treatment groups,the WBC,HBG obviously increased in the Western medicine joint ZQFZ treatment group,(P<0.05). Comparison with the western medicine group,the WBC,PLT close to the ZQFZ treatment group(P>0.05). In HBG, the ZQFZ treatment group was significantly higher than the western medicine group(P<0.05).(2)Comparison of the open field test of mice in each group:After modeling, Compared with the normal control group, crossing and rearing obviously decreased in the model group(P<0.05). After treatment, Compared with the model and the Western treatment groups, the crossing and rearing increased in the ZQFZ treatment and the Western medicine joint ZQFZ treatment groups(P<0.05). Comparison with the Western treatment group, behavior score close to the model group(P>0.05).The ZQFZ Joint western medicine group improve the behavior score more obvious than ZQFZ treatment group(P<0.05).(3)Comparison of the Morris water maze test of mice in each group:After modeling,compared with the normal control group, escape latency increased in the model group(P<0.05). After treatment, the Western medicine joint ZQFZ treatment and the ZQFZ treatment groups escape latency were significantly lower than model group(P<0.05).The escape latency of western treatment group is close to model group(P>0.05). In spatial probe, after successful modeling, model group crossing platform number less normal group, the target area swimming time was significantly shorter than the normal control group(P<0.05).After treatment, the western medicine joint ZQFZ treatment and the ZQFZ treatment groups last number of cross-platform and swimming time in the target area were significantly higher than the model and the western treatment groups(P<0.05).(4)Comparison of the brain tissue in TPH2 m RNA, Ach E m RNA expression of mice in each group:Compared with the normal control group,TPH2 m RNA and Ach Em RNA decreased in the model group(P<0.05). Compared with the model group, TPH2 m RNA and Ach Em RNA increased in ZQFZ treatment and western medicine joint ZQFZ treatment groups(P<0.05). Western treatment group close to model group(P>0.05).(5)Comparison of the hippocampal neuronal morphology of mice in each group: The western treatment and model groups hippocampal CA1,CA3 neurons number significantly reduced, Depigmentation phenomenon is obvious,nerve fibers is sparse and disordered. Nerve nuclei are pyknotic,stain,fragmentation and dissolution. Western medicine joint ZQFZ treatment and ZQFZ treatment groups were close to normal control group,the number of neurous is multiltilayer,form normal, nucleus is round and large,nucleoli is clear.Conclusions:1,After quiet room inhaled benzene(benzene concentration=2000mg/m3)exposure three weeks, the cognition was impaired in mice.2,Zhenqifuzheng capsule can increase blood RT of chronic benzene poisoning mice.3, Zhenqifuzheng capsule can improve cognitive ability of chronic benzene poisoning mice. Which may relate to regulating central neurotransmitters 5-HT,Ach,and relay on protect and repair damaged hippocampal neurons.4, Zhenqifuzheng capsule joint leucogen, batyl alcohol and vitamin B4 not only increase blood RT but also improve cognitive ability of chronic benzene poisoning mice. And it is better than the traditional Chinese medicine and Western medicine alone in improving blood RT.
