Objectives To explore the trophoblast cell surface antigens 2(Trop-2) expression and relationship between Trop-2 expression and clinicopathological factors in colorectal cancer in this study.Methods 84 paraffin embedded colorectal cancer tissue samples and 34 paraffin embedded corresponding tumor-adjacent normal tissue samples were collected from the pathology department of North China University of Science and Technology Affiliated Hospital from July 2009 to October 2012, We detected Trop-2 expressions with Immunohistochemistry and analyzed the relationship between Trop-2 expression and clinicopathological factors of colorectal cancer. 34 removal cancer tissue and corresponding tumor-adjacent normal tissue specimen in radical resection of colorectal cancer from the general surgery and oncology department in October 2012 to October 2014. We also detected removal tissue specimen with Western blot to analyze the Trop-2 expression of colorectal cancer.Results 1. Immunohistochemical staining showed that Trop-2 expression was higher in colorectal cancer tissue samples than that in corresponding tumor-adjacent normal tissues(χ2=8.490)( P<0.05). 2. Colorectal cancer with higher Dukes’ stage(χ2=5.155)(P<0.05) or lymph node metastasis(χ2=3.909)( P<0.05) and distant metastasis(χ2=4.677)( P<0.05), the Trop-2 expression is higher than that in corresponding tissues(P<0.05). There is no significance between Trop-2 expression and gender: male(17/40, 42.5%), female(23/44, 52.3%)(P>0.05), age: ≤50years(11/18, 61.1%), >50years(29/66, 43.9%)(P>0.05), degree of differentiation: well-differentiated(27/49, 55.1%), moderately differentiated(8/23, 34.8%), poorly differentiated(5/12, 41.7%)(P>0.05). 3. Trop-2 expression was also higher in colorectal cancer tissue samples than that in corresponding tumor-adjacent normal tissues in Western blot analysis(P<0.05). The Trop-2 expression is higher in rectal cancer than that in left-sided and right-sided colon cancer(P<0.05), Dukes’ C+D group is higher than Dukes’ A+B group(P<0.05), lymph node metastasis group is higher than no lymph node metastasis group(P<0.05), and distant metastasis group is higher than no distant metastasis group(P<0.05).Conclusions Trop-2 was highly expressed in colorectal cancer, which was closely related to Dukes’ stage, lymph node metastasis, distant metastasis and the tumor site, There is no significance between Trop-2 expression and gender, age, degree of differentiation.
