ObjectiveTo provide molecular genetic basis for oncobiological difference between Left Sided Colon Cancer and Right Sided Colon Cancer, and analyze the differential expressions of Glucose Regulated Protein78kD(GRP78) and Heat Shock Protein20(HSP20) in Left Sided Colon Cancer and Right Sided Colon Cancer.MethodsPathological verified100tissue samples, including Left Sided Colon Cancer and Right Sided Colon Cancer,50respectively were collected and detected for the expressions of GRP78and HSP20by immuno-histochemical SP(Streptavidin Perdcidase) method.Results1.The positive rates of GRP78were78%and32%in colon cancer and pericarcinomatous tissues, respectively. The positive rates in different groups were significantly different (χ2=24.502, p<0.05). The positive rates of GRP78were34%,72%,36%and46%in LSCC, RSCC, left sided pericarcinomatous tissues(LSPT) and right sided pericarcinomatous tissues(RSPT). Chi-square criterion revealed that there were significant differences between the expression in LSCC and RSCC(x2=10.945, p<0.05), LSCC and LSPT(x2=42.747, p<0.05), RSCC and RSPT(x2=8.052, p<0.05), respectively. The expression was correlated with differentiation, invasive depth, lymph node metastasis, liver metastasis and TNM staging (p<0.05), but was not correlated with age, sex and gross morphology(p>0.05).2. The positive rates of HSP20were53%and41%in colon cancer and pericarcinomatous tissues, respectively. The difference in different groups was insignificant(x2=2.890, p>0.05). The positive rates of HSP20were34%,72%,36%and46%in LSCC, RSCC, LSPT and RSPT. Chi-square criterion revealed that there were significant differences between the expression in LSCC and RSCC(χ2=10.945, p<0.05), as well as between RSCC and RSPT(χ2=13.973, p<0.05), respectively. The expression was correlated with gross morphology, lymph node metastasis, TNM staging (p<0.05), but was not correlated with age, sex, invasive depth, differentiation and liver metastasis (p>0.05).3.No signigicant correlation between the expression of GRP78and HSP20in colon cancer tissues was shown by Spearman correlation analysis(r=-0.115, p>0.05).Conclucions1.The expression of GRP78was up-regulated in colon cancer tussues. Compared to RSCC, GRP78was up-regulated in LSCC. GRP78was correlated with differentiation, invasive depth, lymph node metastasis, liver metastasis and TNM staging.2.The positive rates of HSP20between colon cancer and pericarcinomatous tissues was insignificant. HSP20was up-regulated in RSCC. HSP20was correlated with gross morphology, lymph node metastasis, TNM staging.3.There is no correlation between the expression of GRP78and HSP20in colon cancer.4. There were differentially expressed proteins between Left Sided Colon Cancer and Right Sided Colon Cancer, especially for GRP78and HSP20, which were cause by oncobiological difference between Left Sided Colon Cancer and Right Sided Colon Cancer.
