ObjectiveLaryngeal carcinoma is one of the most common malignancies in human headand-neck region. To characterize its molecular mechanisms has been of major interest. In recent studies, several long non-coding RNAs(lnc RNAs) played important roles in the occurrence, development and progress of carcinoma. A few researches revealed that lnc RNAs, acting as oncogenes or tumor suppressors, also expressed aberrantly in head and neck carcinoma. Therefore, there is still no lnc RNA profile of laryngeal carcinoma. It will be of great significances to reveal the molecular mechanism of laryngeal cancer by indentifing the lnc RNA profile of laryngeal cancer. We aim to reveal the lnc RNA profile firstly, then determin the expression level of RP11-169D4.1-001, AC026166.2-001 and its clinical signifcances. Methods1. The global lnc RNA expression profile in LSCC tissues was measured by lnc RNA microarray. GO analysis was performed on the dysregulated lnc RNAs.2. Distinctly expressed lnc RNAs were identified and levels of AC026166.2-001 and RP11-169D4.1-001 lnc RNAs in 87 LSCC samples and paired adjacent normal tissue were analyzed by q RT-PCR.3. PCR sequencing confirmed the PCR product sequence of and RP11-169D4.1-001、 AC026166.2-001.4. We also analyzed the relationship between their expression level of RP11-169D4.1-001, AC026166.2-001 and clinicopathological features. Results1. In this study, 684 lnc RNAs were upregulated and 747 lnc RNAs were downregulated in tumor samples(fold-change >2.0)2. RP11-169D4.1-001 was upregulated by 5.26-fold in LSCC tissues(P=0.002), Conversely,AC026166.2-001 was downregulated by 13.01-fold in LSCC tissues(P=0.008).3. RP11-169D4.1-001 was significantly downregulated in the tumor tissues compared with paired non-neoplastic tissues. Meanwhile, RP11-169D4.1-001 expression was downregulated in metastatic neck lymph nodes compared with paired tumor tissues(P<0.05). We observed significant differences in RP11-169D4.1-001 levels between patients with lymph node metastasis and without(P=0.007). No significant differences were detected between other clinicopathological characteristics.4. We observed that AC026166.2-001 expression was significantly lower in metastatic neck lymph nodes compared with matched normal tissues(P<0.001) and tumor tissues. There was no significant different between AC026166.2-001 levels and age, tumor location, pathological type, lymphatic metastasis and smoking history.5. Our study demonstrated that lower levels of AC026166.2-001 and RP11-169D4.1-001 were significantly correlated with overall survival(P<0.05). ConclusionsTo our knowledge, it’s the first comprehensive lnc RNA profile for LSCC. Both AC026166.2-001 and RP11-169D4.1-001 were identified as novel lnc RNAs with diagnostic value in LSCC, and thus could be potential therapeutic targets. More importantly, according to the survival data analysis, both may be independent factors for prognosis.
