Purpose:To detect the serum levels of retinol binding protein-4 in the short-term (8 weeks) after Roux-en-Y gastric bypass surgery for type 2 diabetic rats, and so as to explore theoretical basis and mechanisms of surgical treatment in type2 diabetes.Methods:The first step is to establish rat mode of the type2 diabetes (SD male) according to our previous study, and then twenty type 2 diabetes rats are divided into two groups by completely randomized method:RYGB surgery group (n=10), sham operation group (n=10)., we take the orbital venous blood to detect blood glucose by blood glucose meter in preoperative and postoperative 1,2,4,8 weeks, and use an enzyme-linked immunosorbent assay (ELISA) kit for determination of serum RBP-4 level. The rats are killed at eight weeks after surgery, and we take rats’ skeletal muscle of the double lower limbs to detect the level of glucose transporter-4 (GLUT-4) by an ELISA kit.Results:There are three died in surgery group and no in sham group; After RYGB surgery, type 2 diabetic rats’ body weight, blood glucose decreased significantly in the surgery group compared with the sham group; Compared with those of sham group, the level of GLUT-4 in the muscle increased significantly and RBP-4 is significantly decreased. There is a negative correlation between RBP-4 and GLUT-4 in skeletal muscle.Conclusions:1.RYGB can decrease the body weight and the blood glucose of type 2 diabetes rats, and the time of decreased blood glucose may hold more longer, indicating the decreased postoperative fasting blood glucose not only relates to weight loss.2.The level of RBP-4 in serum of type 2 diabetes rats is high, and lower the level of GLUT-4 in skeletal muscle. After RYGB surgery, RBP-4 decreased apparently and GLUT-4 in skeletal muscle significantly increased, and RBP-4 negatively correlated with GLUT-4.3.The mechanism of adipokine RBP-4 in the surgical treatment of type 2 diabetes we supposed that RYGB may decrease the level of RBP-4 in serum and increase the level of GLUT-4 in muscle thereby increasing muscle take in glucose,then reducing blood glucose level. |
