[Objective]:To detect the serum levels of retinol binding protein-4 and leptin in the short-time(8 weeks)after duodenal-jejunal anastomosi surgery andduodenal-jejunal bypass surgery for type 2 diabetic rats,and attempt to explore theoretical basis and mechanisms of surgical treatment in type2 diabetes.[Methods]:Twenty non obese type 2 diabetic rats(GK rats)were divided into DJA surgery group(n=10),DJB surgery group(n=10)and sham surgery group(n=10).Weight,food intake,fasting blood glucose,fasting insulin,oral glucose tolerance test(OGTT)were performed,and the area under insulin curve and Homeostasis model assessment-insulin resistant(HOMA-IR)were calculated in preoperative and postoperative,and use an enzyme-linked immunosorbent assay(ELISA)kit for determination of serum RBP-4 and leptin level.The rats are killed at eight weeks after surgery,and we take rats' skeletal muscle of the double lower limbs to detect the level of glucose transporter-4(GLUT-4)by an ELISA kit were detected.[Results]:1.DJB and DJA operation could significantly improve the glucose metabolism and insulin sensitivity of GK rats,which were independent of the weight loss.2.The serum RBP-4 and leptin level were significantly decreased after surgery.The expressions of GLUT4 expressions in theskdetal muscle were signifieantly increased after surgery.[Conclusion]:DJA and DJB operation could significantly improve the glucose metabolism and insulin sensitivity of GK rats,which were independent of the weight loss,but related to the RBP-4/leptin decreased and GLUT-4 significantly increased in skeletal muscle.
