Objective This article investigates the antipyretic effect and intervention mechanism of Kai Qiao Tong Fu method on the dampness-heat of spleen and stomach syndrome through experimental study in the treatment of rat. It expounds the fever control of epidemic febrile disease by brain gut axis, and the relation and actions of brain gut peptides- the material basis and media of brain gut interactions- in the epidemic febrile disease fever. These provide the experimental basis for the treatment of febrile diseases under the guidance of Brain gut interaction theory, and consequently establish the overall concept of pathogenesis and treatment of the brain gut interaction.Methods The experiment references Wu’s molding method and establish fever model of rat dampness-heat of spleen and stomach syndrome. Except the blank and model control groups, the other intervention groups are given drugs of Wang’s lianpu drink, refreshing static injection, big bearing gas soup, paracetamol lavage treatment for 4 groups respectively, Wang’s lianpu drink combining refreshing static injection for 1 group, and Wang’s lianpu drink combining big bearing gas soup for 1 group; all drugs are given by intragastric administration except for refreshing static by caudal vein injection. Recording the temperature change and observing the antipyretic effect. ELISA detecting the content of plasma, hypothalamus, spinal cord, colon, lung, stomach, kidney CRH, AVP, SP, NT, IL-4 and IFN-γ.Analyzing the structural expression of colonic tight junction protein Occludin by immunohistochemistry, gavage of blue dextran-2000 and calculating intestinal propulsive rate.Results The results indicate that treatment groups are as following in antipyretic effect order from best to worst: combining brugs treatment with fast cooling speed and lasting effect, treatment 1, treatment 2, then treatment 3 and treatment 4. There is significant difference in brain gut peptides between the treatment and model groups( P<0.05), and the AVP synthesis of treatment groups are increased which inhibit the production of endogenous pyrogen CRH, so that the content of CRH are reduced and it makes the temperature lowered; while the increase of SP and NT contents also lowers the temperature; and so does the change of IFN-r and IL-4 contents, since the ratio of IFN-r and IL-4 reaches the normal level and it recovers the balance state of TH1/TH2 cells, which are all conducive to Immune regulation and lowering temperature; it also makes thescattered juction protein of colonic Occludin recovered neat and close, enhances intestinal barrier function, slows down the intestinal propulsive rate and restoring of normal intestinal propelling function.Conclusions Both dampness combined Xingnao Kaiqiao method and dampness combined Tongfu Xiere method outperform the traditional dampness methods in treatment of fever rat of dampness-heat of spleen and stomach syndrome. Its Antipyretic Mechanism is relevant to changes of neuropeptide substance CRH, AVP, SP, NT, IL-4, IFN-r content in the rat body. Under the regulation of brain gut axis, it enhances immune function, starts the nerve- endocrine- immune network defense system, so as to regulate body temperature and to maintain homeostasis. The treatment referencing to the overall concept of Chinese traditional medicine and modern medicine theory of brain intestinal connection has obvious advantages for the spleen and stomach damp heat syndrome.
