Objectives:to investigate the effects of ChaiQinTongLin Pian in chronic nonbacterial prostatitis model rats and to explore the underlying mechanisms.Methods: Complete Freund’s adjuvant was injected into the prostate of male rats to establish animal model of chronic nonbacterial prostatitis. Rats were divided into five groups randomly, including the control group, model group, Low-dose group, middle-dose group and high-dose group, Each group of five rats. Two weeks after the injection,1.0g/kg/d、0.5g/kg/d and 0.25g/kg/d suspension of ChaiQinTongLin Pian was intragastric administrated to rats high-, middle- and low-dose group for three weeks respectively, for the control group and model group, same volume of saline was intragastric administrated to rats. Leukyocytes in the expressed prostatic secretion were counted to evaluated the differences among groups. Prostate was derived and paraffin-embedded to obtain specimens and expressed prostatic secretion after administration of five weeks. To dectect pathological changes of prostate after injection, serial sections were obtained for HE staining. S-P immunohistochemical staining was used to investigate the expression of cytokine TNF-a and IL-10. Statistical analysis was performed with the SPSS statistical package(SPSS 22.0).One-way ANOVA was applied for quantitative data of homogeneity of variance and LSD for the pairwise comparisons, nonparametric test was applied for ranked data.A value<0.05 was considered statistical significant.Results:1. Gross and microscope observation of pathological changes:Significant hyperemia and edema were found in prastates from rats that experienced the injection of Complete Freund’s adjuvant. Moreover, microscopic observation of Paraffin sections with H-E stainning showed that prostate glandular cavity shrank from the control group, as well as reduced secretion, atrophic glands and Stromal hyperplasia. Numerous monocytes were found in both connective tissue and gland lumina. Severity of pathological changes was evaluated by rating into 0,Ⅰ. Ⅱ and Ⅲ, there are significant differences among groups(x 2=29.279, P=0.000). The most severe lesions were found in model group rats, followed by low-, middle- and high-dose group. ChaiQinTongLin Pian oral treatment significantly alleviated the lesions caused by injection in the prostate.2. Leukocyte in the expressed prostatic secretion: Different rate of counts of leukocyte were found among groups(x 2=27.319, P=0.000). Much more leukocytes were observed in the model rats’ expressed prostatic secretion. ChaiQinTongLin Pian oral treatment significantly reduced the count of leukocyte in the expressed prostatic secretion.3. TNF-α、IL-10 positive cell count: Significant differences were observed in TNF-α、 IL-10 positive cell count among groups of rats(F=261.487, P=0.000; F=105.374, P=0.000,respectively). The expression of TNF-α-positive cells of model group rat prostate tissue sections was significantly increased compared with the control group (P=0.000), while IL-10-positive cells was significantly decreased (P=0.000). ChaiQinTongLin Pian oral treatment significantly increased the number of IL-10 positive cells and decreased the amount of TNF-α positive cells (P=0.000)Conclusion:Chronic nonbacterial inflammatory lesions in prostate were found in model rats after injection of complete Freund’s adjuvant in the prostate. ChaiQinTongLin Pian oral treatment significantly alleviated the lesions caused by injection in the prostate. The underlying mechanisms could probably related with the reduction of expression of proinflammatory cytokine such as TNF-α and the increased expression of anti-inflammatory factor of IL-10.
