Objective: To explore the change of inflammatory process and inflammatory-related factors and observe the effect of Chinese Herbs for Activating Blood Circulation on modeling rats. Methods: Male rats model were established by abdominal intradermal multiple points injection twice with different concentration of saline extract of male rat prostate glands (RPG) in Freund's complete adjuvant (FCA) within 30 days and the injection of Pertussis-Diphtheria-Tetanus vaccine into the abdominal cavity. The prostate tissue was observed to evaluate whether the models was successful. The indexes such as COX-2, T-AOC, NO and PGE2 in prostate tissue and serum were determined to analyze their correlation. After 4 weeks, the modeling rats treated with single and compound high, middle and low-dosage Chinese Herbs for activating blood circulation were sacrificed. Results: (1) In chronic nonbacterial prostatitis model groups treated with saline extract of male rat prostate glands with concentration of 20mg/m1, 40mg/m1 and 80mg/ml 45 days later, interstitial was full of lymphocytes infiltration, it showed significantly chronic inflammation changes with concentration-dose effect compared with control group (P<0.05). (2) In chronic nonbacterial prostatitis model group treated with 60mg/m1 saline extract, it showed significant different in lymphocytes infiltration and interstitial proliferation with time-dose effect. Compared with control group, lymphocytes infiltration and interstitial proliferation showed significant difference in 6 and 8-week groups (P<0.05). NO concentration in serum compared with control group(P<0.05). NO in prostate tissue increased significantly in 2, 4, 6 and 8-week groups(P<0.05). T-AOC decreased significantly in 4, 6 and 8-week groups (P<0.05). Prostaglandin E2 in serum increased significantly compared with control group(P<0.05). (3) In Chinese Herbs intervention groups, compared with model group, lymphocytes infiltration and interstitial proliferation showed significant difference in compound high, middle-dosage and single groups. NO concentration in serum decreased significantly in compound high, middle-dosage and single groups(P<0.05). NO in prostate tissue compound high, middle-dosage and single groups(P<0.05). T-AOC in prostate tissue increased significantly in compound high-dosage and single groups(P<0.05). Prostaglandin E2 in serum increased significantly in compound high-dosage group and decreased significantly in single group compared with model group(P<0.05). Conclusion: It was an effective method for establishing the chronic nonbacterial prostatitis model in Wistar rats by intracutaneous injection in multi-loci of RPG mixed with FCA and peritoneal injection with Pertussis-Diphtheria-Tetanus vaccine. High, middle dosage compound and single Chinese herbs could reduce the prostatic inflammatory response and fibrosis severity of model rat and protect the prostate tissue.
