Obejective:Observe the anti-inflammatory effect and mechanism of moxibustion therapy on "Shen Shu"and "Zu sanli"on RA model of SD rats and with the method of the IL-17A lentivirus-mediated model.The aim is to provide much more experimental basis for clinical application of moxibustion treatment of RA.Method:Separate 54 rats into 6 groups:blank group、blank group with moxibustion、RA group、 RA moxibustion group、IL-17 overexpression group and IL-17 overexpression with moxibustion group. Treat relative group with festering moxibustion with 3 courses.Each course lasts 6 days,resting 1 day between two courses. Record the data variation of the behavioristic RA rats of the arthritic flexibility score and foot swollen volume. Test the concentration of IL-1b、TNF-a、 IL-17A in the peripheral blood by ELISA and the IL-17-mRNA by PCR and the IKβ-zeta by Western blot.Result:1 The arthritic flexibility score and foot swollen volume both increase after the model being established, while after the treatment of festering moxibustion, these two score both decrease.2①By comparison with blank group,the RA rats groups has the concentration of IL-1b、 TNF-a、IL-17A increase in the peripheral blood obviously (P< 0.01);after treatment of festering moxibustion,there is no significant difference between treatment group and blank group (P> 0.05) comparing RA and RA moxibustion group, RA moxibustion group has lower concentration of IL-1b、TNF-a、IL-17 and IL-17-mRNA(P<0.01);②By comparison with blank group,the RA rats groups has lower the activity of IKβ-zeta protein (P< 0.01);after treatment of moxibustion,there is no significant difference between blank group and blank group with moxibustion of the activity of IKB-zeta (P>0.05);comparing RA and RA moxibustion group, RA moxibusion group has lower concentration of IL-lb、TNF-a、IL-17 and IL-17-mRNA(P<0.01); comparing RA and RA moxibustion group, RA moxibustion group has higher activity of IKB-zeta.3 ①By comparing IL-17 overexpression group and RA group, IL-17 overexpression group has the increase of the concentration of IL-lb、TNF-a、IL-17A and IL-17-mRNA(P<0.01); IL-17 overexpression with moxibusion group has great decrease of the concentration of IL-1b、TNF-a、 IL-17A and IL-17-mRNA(P<0.01) with the comparison with IL-17 overexpression group.②By comparing IL-17 overexpression group and RA group, IL-17 overexpression group has lower activity of IKβ-zeta; IL-17 overexpression with moxibusion group has higher activity of IKβ-zeta.Conclusion1. The arthritic flexibility score can be decreased and foot swollen volume can be alleviated both by the therapy of moxibustion, indicating that moxibustion has the effect of anti-inflammation and analgesia in RA rats.2. ①he concentration of IL-lb、TNF-a、IL-17A and IL-17-mRNA can be decreased by moxibustion, implying that the mechanism of moxibustion in treating RA are achieved by decrease inflammatory cytokine to alleviate the synovitis and to improve the local syndrome. ② Moxibustion can increase the activity of IKP-zeta,hinting that moxibustion can increase the anti-inflammation effect of IKβ-zeta.3. ① The concentration of IL-lb、TNF-a、IL-17A and IL-17-mRNA of IL-17 overexpression group can be decreased by moxibustion, pointing that the anti-inflammation effect of moxibustion is attained by the suppression of IL-17 which in turn decrease the level of inflammatory cytokine IL-lb、TNF-a in blood.②Moxibustion can increase the activity of IKβ-zeta in IL-17 overexpression group, reminding that moxibustion has the capability of increase the activity of IKβ-zeta by suppressing the release of IL-17.
