This thesis mainly deals with chemical constituents from Ganoderma cochlear and their renoprotective effects. Among them, aromatic meroterpenoids constitute a major part.Lingzhi, a superior medicine listed in "Shen Nong Ben Cao Jing", also known as the mushroom of immortality, is a valuable traditional Chinese medicine. In recent years, scientific studies have found that the main chemical constituents of the genus Ganoderma consisted of triterpenes, polysaccharides, steroids, polypeptides, alkaloids, flavonoids, fatty acids and other constituents. Especially, triterpenes and polysaccharides have been considered as the main chemical constituents for long time. Previous studies also found that Ganoderma species contained meroterpenoids. In order to discovery more meroterpenoids from the genus Ganoderma,46 compounds were isolated from G. cochlear, by the means of chromatographic methods (MCI gel CHP 20P, Sephadex LH-20, RP-18, HPLC, and PTLC) and spectroscopic evidences (1D and 2D NMR, MS, UV). Among them,25 compounds were new ones, including 19 new aromatic meroterpenoids (compounds 1-12 and 19-25) as well as 6 new aikaloids (13-18). Furthermore, The known compounds were identified as ganomycin C (26), fornicin B (27), ganomycin I (28), fornicin C (29), ganocochlearin A (30), ganocochlearin C (31), daldinone A (32), fornicatin G(33), hydroquinone (34), cyclo(Pro-Leu) (35), cyclo[(D)-Pro-(D)-Phe] (36), aesculetin (37),3-methoxy-4-hydroxyphenylethanol (38),4-hydroxy acetophenone (39), methyl p-hydroxy-cinnamate (40),2-methoxy-4-hydroxybenzaldehyde (41),4-hydroxy-3-methoxy benzoic acid (42), 2-hydroxy-5-ethoxybenzoic acid (43),2-(2,5-dihydroxyphenyl)acetic acid (44), 1-phenylethane-1,2-diol (45) and (E)-5-(2,5-dihydroxyphenyl)pent-4-en-2-one (46). It is worth mentioning that all compounds possessing chiral centers are racemic, and we achieved the separation of all racemes by using chiral materials. For part of the above compounds, we carried out the bioactive study around the related vitro model of chronic kidney disease, and found that some compounds have anti-renal fibrosis.This research will provide important data of the meroterpenoids from the genus Ganoderma.
