Objective: Ganoderma cochlear, one of the most famous traditional Chinese medicinal mushrooms, is a popular folk and oriental medicine used to treat many diseases. As the scientific study has found, the main chemical constituents of Ganoderma lucidum is triterpenes、polysaccharide、steroids、protein、polypeptide、alkaloids、flavonoids、fatty acids and other compounds. These compounds have very good regulatory effect on human immune system, nervous system, cardiovascular system. However, the current research focused on triterpenes and polysaccharide, and about the trace alkaloids research is so less. But we believe that the tarce alkaloids chemical constituent must have very important physiological activity. In consequence, this topic aimed at further more research of alkaloid constituents in Ganoderma cochlear.Methods: The powdered fruiting bodies of Ganoderma cochlear(100 kg) were extracted with 70% Et OH at room temperature and concentrated under reduced pressure(6 × 300 L × 48 h) to give a crude extract(10 kg), which was suspended in water followed by extracted with ethyl acetate to afford an ethyl acetate soluble extract(2 kg). The compounds were isolated by combining the traditional and modle method(include silica gel, MCI gel CHP 20 P, Sephadex LH-20(Me OH), RP-18 column chromatography, HPLC, and Preparative TLC). Their structures of compounds were elucidated on the basis of phsiochemical characteristics and spectral methods such as UV, 1H NMR, 13 C NMR, DEPT, HSQC, HMBC,1H-1H COSY and so on.Results: Twenty four compounds were obtained from ethyl acetate fraction of the extract from Ganoderma cochlear,twenty four compounds were elucidated by spectroscopic and chemical methods, as ganocochlearine A(1), ganococh learine B(2), 3-13,(±)-sinensilactam A(14), 15-17,sinensine E(18), ganomycin I(19),fornicin B(20),2,3-dihydro-4(1H)-quinolone(21), 8-hydroxy-2,3-dihydro-4(1H)-quinolone(22), fornicatin A(23) and(3S,6S)-3-[(1′S)-1-methylpropyl]-6-(phenyl methyl)-piperazine-2,5-dione(24).(The compounds 3-13 and 15-17 don’t be na med for no publish.)Conclusion: 14 new alkaloids were obtained from Ganoderma cochlear revealing a new insight into the chemical profile of this species, provide reference on effcient materials for further research of Ganoderma.
