Objective:Construction of ischemia/reperfusion model to explore gastrodin inhibiting autophagy in order to achieve the cardioprotective effect and mechanism of action in vivo and vitro experiments.Methods:In vivo:The 80 SD adult male rats (200-300g) were randomly divided into 10 groups (n=8):control group (continuous perfusion with KH solution 90min), ischemia-reperfusion group (after global ischemia 30min, restore perfusion 60min), the treatment group after treatment gastrodin (restore perfusion and perfusion 30min gastrodin each 20,50,100,200μM), the application of retrograde Langendorff isolated heart perfusion model to measure cardiac function LVDP, LVEDP, dp/dtmax,-dp/dtmax, CF, HR change; the use of TTC staining of myocardial infarct size; Immunohist ochemical with since confocal microscopy detection the expression of autophagy-related protein LC3.In vitro:Using serum deprivation-induced myocardial ischemia model in H9c2 cell, H9c2 of rats cardiomyocytes cultured were randomly divided into control group, different time (0,3,6,12,24,48h) serum deprivation group, different time (0,6,12,24h) serum deprivation + gastrodin (20μM) post-treatment group and positive control group. Using Western blotting analysis to detect the expression of autophagy-related factor LC3, Atg5, Beclin 1; Immunofluorescence double labeling method to detect the expression changes of autophagy protein LC3.Results:In vivo:Ischemia/reperfusion reduces cardiac function of LVDP, LVEDP,+ dp/dtmax,-dp/dtmax, CF, increase HR, gastrodin can improve these indicators, in a concentration- dependent manner. Gastrodin can improve myocardial infarct size. Gastrodin can improve the expression of ischemia/reperfusion induces autophagy-related factors LC3.In vitro:Serum deprivation can induce H9c2 cardiomyocyte upregulated autophagy-related factors LC3, Atg5, Beclin 1, in a time of gradient; gastrodin can inhibit the expression of serum deprivation-induced autophagy-related factor LC3, Atg5, Beclin 1 in H9c2 cardiomyocytes.Conclusion:Gastrodin can inhibit autophagy in the myocardium, thereby inhibiting myocardial ischemia-induced injury in order to protect the myocardium.
