Objective To investigate the expression of Th17 cell-associated factor IL-17 and phosphorylated S6 ribosomal protein (p-S6) which is a target protein downstream of the mTOR signaling pathway in skin lesions tissue of pemphigus vulgaris.To explore the activation state of the mTOR signaling pathway in skin lesions tissue of PV and provide new targets and approaches for the treatment of pemphigus vulgaris.Methods skin lesions tissue specimen were obtained from 12 patients of PV(PV group)with acute exacerbation in the department of Dermatology,the Second Affiliated Hospital of Kunming Medical University.To collect skin samples of healthy people of Plastic Surgery as a control group, excluding PV and other autoimmune diseases.Test the expression of IL-17 and p-S6 in the fresh lesions of PV and Skin tissue of the healthy control group by immunofluorescence technique. The result and the images were required by Fluorescence microscope (OLYMPUS BX51, JAPAN) and Microscopic imaging software cellsens.The two groups were respectively compared. Assess the amount aid IPP (Image-Pro Plus 6.0) software, the results are expressed as integrated optical density (IOD) values.Using the SPSS 17.0 software package to analysis the results. Measurement datas were distributed for mean ±SEM. two groups were compared using t’test. Correlation analysis between variables were analyzed using Pearson Linear correlation analysis. P<0.05 were regarded as statistically significant.Result The expression levels (Represented by the integrated optical density value)of IL-17 and p-S6 in the tissue lesions of PV, the results were respectively(5628.697 ± 1941.51448) and (4612.835 ± 2059.84898),while the normal control group were respectively (3864.8175 ± 692.66423) and (2583.2863 ± 242.2068). The difference was statistically significant between the two groups (P <0.05). The expression levels of IL-17 and p-S6 present correlation (r=0.581,P <0.05).Conclusion1. IL-17 is highly expressed in skin lesions tissue of pemphigus vulgaris, IL-17 may be involved in the pathogenesis of pemphigus.2. p-S6 is highly expressed in skin lesions tissue of pemphigus vulgaris, activation of mTOR signaling pathway may be involved in the pathogenesis of pemphigus.3. The expression levels of L-17 and p-S6 present correlation.
