Purpose:Exploring the effect of Xuebijing injection to immune regulating and the possible mechanism through the transcription factor T-bet and GATA-3 of T cells in patients with sepsis.Method:This research adopted the method of prospective study.These sepsis patients that were collected in emergency intensive care unit (ICU) from June to December in 2014 are randomly divided into treatment group and control group.The control group and treatment group respectively collected eleven sepsis patients.Six people were included in the control group and nine people were included in the treatment group at last.In the ICU the control group were given the traditional treatment including the fighting infection、adding the liquid、correcting electrolyte disorder、nutritional support、treating the primary disease and so on.The treatment group were additionally given Xuebijing injection 100ml twice a day and the Xuebijing injection must be continuously used at least seven days.Recording the basic information such as age、gender and the APACHE II score of the first and seventh day.Collecting 4ml peripheral venous blood for every sepsis patients on the Od(prior treatment)、1d、3d and 7d after entering the ICU.Two milliliter blood was collected in EDTA tube for the inspection of T-bet mRNA and GATA-3 mRNA that respectively were the specific transcription factors of Thl and Th2 in the method of RT-PCR.Two milliliter blood was collected in the coagulant tube for the inspection of IFN-y and IL-4 concentration.Judging the function of Xuebijing injection to immune regulating in sepsis patients through IFN-γ、IL-4、IFN-y/IL-4、T-bet mRNA、GATA-3 mRNA and T-bet mRNA/GATA-3 mRNA.Evaluating the effect of Xuebijing injection to sepsis patients through APACHE Ⅱ score.Results:1、There was no statistically difference between the treatment group and control group in age、gender and the APACHE Ⅱ score on the first day.2、There was no statistically difference between the treatment group and control group in T-bet mRNA ' GATA-3 mRNA on the 0d、Id and 3d. When the treatment group compared with the control group on the 7d, T-bet mRNA was higher and GATA-3 mRNA was lower.There was statistically significant.3、There was no statistically difference between the treatment group and control group in T-bet mRNA/GATA-3 mRNA on the 0d、1d and 3d.The T-bet mRNA /GATA-3 mRNA in treatment group increased higher compared with the control group on the 7d and there was statistically significant.4、There was no statistically difference between the treatment group and control group in IFN-y and IL-4 on the 0d、1d and 3d. When the treatment group compared with the control group on the 7d,IFN-γ was higher and IL-4 was lower.There was statistically significant.5、There was no statistically difference between the treatment group and control group in IFN-γ/IL-4 on the Od. Id and 3d.The IFN-γ/IL-4 in treatment group increased higher compared with the control group on the 7d and there was statistically significant.6、The APACHE Ⅱ score significantly decreased compared with the control group on the 7d and there was statistically difference.Conclusion:1、The Xuebijing injection can enhance the immune function and decrease the drifting from Thl to Th2 through increasing T-bet mRNA/GATA-3 mRNA for sepsis patients.2、The Xuebijing injection can promote the recovery of pro-inflammatory and anti-inflammatory balance through increasing the IFN-γ/IL-4 for sepsis patients.3、The Xuebijing injection can decrease the APACHE Ⅱ score and severity for sepsis patients.
